It was stirred further for 20 minutes at room temperature at which time LC/MS analysis confirmed full consumption of starting material. Pure compound ((lR,3s,5S)-3-(3-(6-(2-hydroxypropan-2- yl)pyridin-3-yl)-8H-pyrazolo[l,5-a]pyrrolo[3,2-e]pyrimidin-5-yl)-8-azabicyclo[3.2.1]octan-8- yl)(lH-l ,2,4-triazol-3-yl)methanone was isolated by preparative HPLC.

The resulting mixture was stirred for 30 minutes then concentrated under reduced pressure. 1H-[1,2,3]Triazole-4-carboxylic acid (272 mg, 2.4 mmol) and HATU (959 mg, 2.5 mmol) were combined in DMF (2 mL) and stirred for 10 minutes. DIPEA (1.3 mL, 7.2 mmol) and Compound 1 in DMF (2 mL) were added and the resulting mixture was stirred for 30 minutes then concentrated. The residue was purified by reverse phase chromatography (20-100% MeCN in water) to yield the title compound (287 mg).

The organics were washed with brine, dried over magnesium sulfate, filtered, and concentrated. The crude material was purified by silica gel gradient chromatography (0-100% ethyl acetate in hexanes), providing the titled compound as a yellow-white solid. LRMS m/z (Mu+Eta) 287.3 found, 287.1 required.

The aqueous phase was neutralized to pH 7, and further extracted with EtOAc. The combined organics were concentrated in vacuo. Purification via flash column chromatography (EtOAc/Hexanes) yielded the title compound as an off white solid (3.78 g, 73percent): mp 67-69° C.; 1H NMR (400 MHz, CDCl3) delta 8.43 (s, 1H), 7.70 (m, 2H), 7.38 (m, 2H); 19F NMR (376 MHz, CDCl3) delta -58.02.
