The reaction mixture was stirred for 5 h. After water and EtOAc were added, the organic layer was extracted with EtOAc. The organic layer was washed with water and brine, dried over anhydrous Na2SO4, filtered off, and concentrated in vacuo. The crude product was purified by prep HPLC. The pure fractions were lyophilized yielding (S)-4-(2-amino-5-(1-methyl-1H-1,2,3-triazol-4-yl)pyridin-3-yl)-N-(1-(3-chlorophenyl)-2-hydroxyethyl)-2-fluorobenzamide as a TFA salt (25% over 2 steps). LCMS (m/z): 467.3 (MH+), 0.61 min; 1H NMR (400 MHz, CD3OD) delta ppm 8.64 (m, 1H), 8.31 (m, 1H), 8.25 (m, 2H), 8.2 (m, 1H), 7.82 (m, 1H), 7.4 (m, 3H), 7.28 (m, 2H), 7.22 (m, 1H), 5.11 (m, 1H), 4.07 (s, 3H), 3.78 (m, 2H).

A mixture of 2-(2H-1,2,3-triazol-2-yl)benzoic acid (50 mg, 0.26 mmol) in thionyl chloride (2 mL) was stirred at 80C for 1 h, cooled to RT and concentrated in vacuo. The residue was dissolved in DCM (10 mL) and added to a solution of the product from step 5 (66 mg, 0.26 mmol) in DCM (1 mL) and DIPEA (0.1 ml) at 0C. The resulting mixture was stirred at RT for 12 h, poured into water and extracted with EtOAc (50 mL x 3). The combined organic layers were washed with brine, dried over MgSO4, filtered and concentrated in vacuo. The crude product was purified by Prep-TLC (50% EtOAc in petroleum ether) to give the title compound as a white solid.

The reaction mixture was heated at 115 °C for 12 h. The reaction mixture was then evaporated to dryness using toluene as an azeotropic solvent and triturated with diethyl ether. This was finally dried under high vacuum which afforded 66b as a deep brown solid (300 mg, 40percent). This was then used in the next step without any further purification. LCMS(ESI) m/z 288.02 [M-H+]; 57percent (purity).

After being stirred for 18 h at r.t., the reaction mixture is filtered through celite and concentrated under reduced pressure. The residue is diluted with AcOEt and filtered again. The filtrate is concentrated under reduced pressure to give the crude imine. To a solution of the crude imine in 12 ml of MeOH, 0.756 mg (20 mmol) of NaBH4 is added at -10 C. After being stirred for 1 h, the reaction is quenched with sat. NH4Cl and the reaction mixture is extracted with AcOEt. The combined extracts are washed with brine, dried over MgSO4 and concentrated under reduced pressure. The residue is purified by column chromatography on silica gel (eluent: n-Hexane: AcOEt=1:10) to give the title compound. 1H-NMR (400 MHz, CDCl3): 1.29 (t, 3H, J=7.04 Hz), 1.90 (brs, 1H), 3.43 (s, 2H), 3.88 (s, 2H), 4.21 (q, 2H, J=7.04 Hz), 7.49 (d, 2H, J=8.56 Hz), 7.64 (d, 2H, J=8.56 Hz), 8.10 (s, 1H), 8.55 (s, 1H).

The reaction mixture was irradiated by 400 W microwave at 80 oC, for the specified reaction times. TLC with n-hexan:ethylacetate eluent was used to monitor the progress of the reactions. After reaction completion and cooling, the reaction mixtures were filtered and washed with ethanol to separate the catalyst. First, 10 mL water was added to the solution. Then, EtOH/H2O (4:1) was added to crystallize the water insoluble crude products. Then the crystals were filtered to achieve pure products (4a-h).

The resulting solid was triturated with diethyl ether. The solid was collected by filtration and washed again with diethyl ether to afford 1-trityl-1H-[1,2,4]triazole-3-carboxylic acid methyl ester as a white solid (2.45 g, 84percent) which was used without further purification: LR-FAB for C23H19N3O2 (M+) at m/z=369.
