Peroxisomes are metabolically active organelles which are known to exert anti-inflammatory effects especially associated with the synthesis of mediators of inflammation resolution. However, the role of catalase and effects of peroxisome derived reactive oxygen species (ROS) caused by lipid peroxidation through 4-hydroxy-2-nonenal (4-HNE) on lipopolysaccharide (LPS) mediated inflammatory pathway are largely unknown. Here, we show that inhibition of catalase by 3-aminotriazole (3-AT) results in the generation of peroxisomal ROS, which contribute to leaky peroxisomes in RAW264.7 cells. Leaky peroxisomes cause the release of matrix proteins to the cytosol, which are degraded by ubiquitin proteasome system. Furthermore, 3-AT promotes the formation of 4HNE-I kappa B alpha adduct which directly interferes with LPS induced NF-kappa B activation. Even though, a selective degradation of peroxisome matrix proteins and formation of 4HNE- I kappa B alpha adduct are not directly related with each other, both of them are could be the consequences of lipid peroxidation occurring at the peroxisome membrane.

For the first time, the coordination-based energy sacrificial bonds have been constructed in the interface between lignin and polyolefin elastomer for preparing a new class of high performance thermoplastic elastomers (TPEs) with biomass lignin as the hard plastic phase. The coordination bonds not only promoted the dispersion of lignin, but also improved the interfacial interactions between lignin and polyolefin elastomer matrix, and also facilitated the orientation of chain segments during stretching. The synergistic coordination effect of lignin promoted higher energy dissipation, leading to simultaneously enhanced strength and toughness of lignin-based TPEs even with the lignin loading as high as 30 wt %. The lignin-based TPEs also exhibited excellent shape memory performance. Our strategy offers a promising methodology for the facile production of high performance but cost-effective TPE materials using biorenewable resources as plastic phase.

Human activities such as agriculture, aquaculture, and industry can lead to the pollution of coastal waters by trace organic contaminants (TrOCs), and the TrOCs can pose a threat to marine ecosystems. Therefore, it is essential to investigate the occurrence, distribution, and ecological risk of the TrOCs in coastal waters. Previous studies adopting conventional analytical methods have focused on a limited number of targets. Herein, a comprehensive and systematic determination was undertaken to target 484 TrOCs in the waters around the Liaodong Peninsula, China. Eighty-six TrOCs were detected at concentrations of up to 350 ng L-1, and 25 TrOCs were detected at a frequency of >50%. Pesticides were the predominant pollutants, occurring at high concentrations with large detection frequencies. Ecological risks were assessed for single pollutants and mixtures based on the risk quotient and concentration addition modeling, respectively. The detected pesticides posed relatively high risk to aquatic organisms, while pharmaceuticals, consumer products, and other pollutants posed little or no risk. TrOC mixtures posed extremely high risk to aquatic organisms, which represented a significant threat to the marine environment and local communities. The results described here provide useful information that can inform China’s Action Plan for Prevention and Control of Water Pollution. (C) 2020 Elsevier Ltd. All rights reserved.

Environmental pollution by the heavy usage of pesticides has been a pandemic issue in view of the rising farming operations for increasing the crop yield to meet the requirements of food chain supply. Throughout the world, environmental pollution by the presence of pesticides, particularly the use of herbicides in large quantities to protect the crops, has posed many environmental issues. In this research, an electrochemical sensor based on tungsten oxide hydrates (WO3 center dot 0.33H(2)O) nanorod modified carbon paste electrode (CPE) was developed for the detection of herbicide, amitrole (AMT) by the cydic voltammeter. Hydrotheimally synthesized and characterized WO3 center dot 0.33H(2)O nanorod was found to be sensitive towards the detection of AMT due to its superior sensing property as the sensor showed enhanced current and catalytic property when used in phosphate buffer solution (PBS) of pH 5.0 by the cyclic voltammetric (CV) and square wave voltammetric (SWV) techniques. The influence of electro kinetic parameters viz., scan rate, pH, accumulation time and temperature with respect to AMT oxidation was studied using CV. The linearity range was in between 1.0 x 10(-8) M and 24 x 10(-5) M and limit of detection (LOD) and limit of quantification (LOQ) was calculated to be 233 nM and 7.8 nM respectively. The proposed simple method demonstrated the potential applicability to detect AMT from the soil and water samples. (C) 2020 Elsevier B.V. All rights reserved.

Visualization of intracellular transport pathways is crucial to investigate the internalization mechanism and understand the intracellular behavior of nanomaterials. Herein, we rationalized the design of micellar nanopartides (NPs for ratiometric fluorescent mapping of intracellular pH and glutathione (GSH), two essential parameters for maintaining normal cellular functions. Specifically, pH-sensitive naphthalimide-based probe (NPI) and pH-inert rhodamine B (RhB) were covalently labeled to double hydrophilic block copolymers (DHBCs) using the thiolactone chemistry, enabling the covalent attachment of NPI and RhB to one molecule with a redox-responsive disulfide linkage. The dually labeled DHBCs exhibited blue/orange dual emissions in acidic pH, which was further converted into green/orange dual emissions in neutral pH because of the deprotonation of NPI moieties and the sole green emission in the presence of GSH at neutral pH because of the decreased Forster resonance energy transfer efficiency between an NPI donor and an RhB acceptor as a result of GSH-mediated cleavage of disulfide bonds. These remarkable ratiometric fluorescence changes allowed for not only the simultaneous mapping of the intracellular pH and GSH but also the intracellular transport pathways of internalized NPs.

In vitro genotoxicity testing that employs metabolically active human cells may be better suited for evaluating human in vivo genotoxicity than current bacterial or non-metabolically active mammalian cell systems. In the current study, 28 compounds, known to have different genotoxicity and carcinogenicity modes of action (MoAs), were evaluated over a wide range of concentrations for the ability to induce DNA damage in human HepG2 and HepaRG cells. DNA damage dose-responses in both cell lines were quantified using a combination of high-throughput high-content (HTHC) CometChip technology and benchmark dose (BMD) quantitative approaches. Assays of metabolic activity indicated that differentiated HepaRG cells had much higher levels of cytochromes P450 activity than did HepG2 cells. DNA damage was observed for four and two out of five indirect-acting genotoxic carcinogens in HepaRG and HepG2 cells, respectively. Four out of seven direct-acting carcinogens were positive in both cell lines, with two of the three negatives being genotoxic mainly through aneugenicity. The four chemicals positive in both cell lines generated HTHC Comet data in HepaRG and HepG2 cells with comparable BMD values. All the non-genotoxic compounds, including six non-genotoxic carcinogens, were negative in HepaRG cells; five genotoxic non-carcinogens also were negative. Our results indicate that the HTHC CometChip assay detects a greater proportion of genotoxic carcinogens requiring metabolic activation (i.e., indirect carcinogens) when conducted with HepaRG cells than with HepG2 cells. In addition, BMD genotoxicity potency estimate is useful for quantitatively evaluating CometChip assay data in a scientifically rigorous manner.

Filamentous fungi Monascus sp. has been utilized for fermentative production of food colorant (Red Yeast Rice) for more than 1000years in China. The main colorant components of Red Yeast Rice are mixture of red Monascus pigments (RMPs) with various primary amine residues. In the present work, the non-natural primary amine p-aminobenzamide, exhibiting as non-involved in nitrogen microbial metabolism, nontoxicity to microbial cells, and chemical reactivity with orange Monascus pigments (OMPs), was screened. Based on the screened result, RMPs with the single p-aminobenzamide residue were produced by cell suspension culture in a nonionic surfactant micelle aqueous solution via in situ chemical modification of OMPs. Furthermore, in situ chemical modification of OMPs also provided a strategy for maintaining a relatively low OMP concentration and then efficient accumulation of high concentration of RMPs (3.3g/l).

Intracerebroventricular (icv) injection of hydrogen peroxide (H2O2), a reactive oxygen species, or the blockade of catalase (enzyme that degrades H2O2 into H2O and O-2) with icv injection of 3-amino-1,2,4-triazole (ATZ) reduces the pressor effects of angiotensin II also injected icv. In the present study, we investigated the effects of ATZ injected icv or intravenously (iv) on the pressor responses induced by icv injections of the cholinergic agonist carbachol, which similar to angiotensin II induces pressor responses that depend on sympathoexcitation and vasopressin release. In addition, the effects of H2O2 icv on the pressor responses to icv carbachol were also tested to compare with the effects of ATZ. Normotensive non-anesthetized male Holtzman rats (280-300 g, n = 8-9/group) with stainless steel cannulas implanted in the lateral ventricle were used. Previous injection of ATZ (5 nmol/1 mu l) or H2O2 (5 mu mol/1 mu l) icv similarly reduced the pressor responses induced by carbachol (4 nmol/1 mu l) injected icv (13 +/- 4 and 12 +/- 4 mmHg, respectively, vs. vehicle + carbachol: 30 +/- 5 mmHg). ATZ (3.6 mmol/kg of body weight) injected iv also reduced icv carbachol-induced pressor responses (21 +/- 2 mmHg). ATZ icv or iv and H2O2 icv injected alone produced no effect on baseline arterial pressure. The treatments also produced no significant change of heart rate. The results show that ATZ icv or iv reduced the pressor responses to icv carbachol, suggesting that endogenous H2O2 acting centrally inhibits the pressor mechanisms (sympathoactivation and/or vasopressin release) activated by central cholinergic stimulation.

Eight energetic salts with a gem-dinitro group based on 1,2,4-oxadiazole were developed. All of the structures were confirmed by infrared (IR) spectroscopy, H-1 and C-13 nuclear magnetic resonance (NMR) spectroscopy, and high-resolution mass spectrometry (HRMS), and single-crystal X-ray diffraction was used to determine the structures of the pivotal intermediate 3-(chlorodinitromethyl)-5-methyl-1,2,4-oxadiazole (3), the potassium salt (4) and the hydrazinium salt (7). All of them are insensitive to impact (>40 J) and have higher energy levels than trinitrotoluene (TNT).

A new series of indole containing biaryl derivatives were designed and synthesized, and further biological evaluations of their antiproliferative activity against cancer cell lines (MGC-803 and TE-1 cells) were also conducted. Of these synthesized biaryls, compound 4-methyl-2-((5-methyl-[1,2,4]triazolo[1,5-a]pyrimidin-7-yl) methyl)quinazoline (23) performed as the most potent antiproliferative agent that inhibited cell viability of MGC-803 cells with an IC50 value of 8.28 mu M. In addition, investigation of mechanism exhibited that the compound 4-methyl-2-((5-methyl-[1,2,4]triazolo[1,5-a]pyrimidin-7-yl)methyl)quinazoline (23) could inhibit the expression of c-Myc and glycolysis related proteins, decrease the ATP and lactate production, and further induce apoptosis by activating the AMP-activated protein kinase (AMPK) and p53 signaling pathways.
