Recently, activated carbon adsorption for water treatment regained substantial attention due to the emerging task to remove trace organic compounds such as pesticides. In many applications, especially in decentralized water treatment, one major drawback of adsorbents is their limited recyclability due to inadequate logistics or uneconomical reactivation. In this lab-scale study, we present the temperature swing adsorption in the aqueous phase that allows the in situ regeneration of fixed-bed adsorbers, and prove its technical feasibility. Complying with circular water economy principles, we eliminated the pivotal need for regular replacement and consumables by employing only clean water instead of dedicated regeneration solutions. Adsorption of the herbicide amitrole in aqueous solution on granular activated carbon was exothermic (Delta H = -14.4 +/- 3.2 kJ mol(-1) for T = 20-94 degrees C) and followed the Freundlich model. The proposed method consisting of a short counterflow flush with liquid water at 125 degrees C effectively regenerated the adsorbent. Hence, we obtained a cyclic steady state operation with breakthrough after 122 +/- 14 bed volumes (at c(out)/c(in) = 0.2), cycle-average rejection of 90 +/- 1%, and water recovery of up to 78 +/- 4%. No thermal aging of adsorbent was observed over the investigated 17 cycles.

Solvothermal reactions of Zn(NO3)(2)center dot 6H(2)O, 3-amino-1,2,4-triazole as well as three different carboxylate ligands afforded three novel coordination polymers: [Zn-2 (azdc) (atz) (OH) (H2O)](n) (1), {[Zn-1.5 (btc)(0.5) (atz)]center dot 1.5H(2)O}(n) (2), and {[Zn-4 (nipa)(2) (atz)(3)(OH) (H2O)]center dot 2H(2)O}(n) (3) (H(2)azdc = 4,4′-azobenzoic acid, H(4)btc = 1,2,4,5-benzenetetracarboxylic acid, 5-H(2)nipa = 5-nitroisophthalic acid, Hatz = 3-amino-1,2,4-triazole). Moreover, the three coordination polymers have been characterized by IR spectroscopy, elemental analysis, single-crystal X-ray diffraction, and powder X-ray diffraction techniques. Complex 1 reveals a 2D layer structure, which is further connected through pi-pi stacking interactions forming a 3D supramolecular network. Complex 2 exhibits a 3D porous framework with 1D open channels. Complex 3 shows a 3D pillared-layer framework. Notably, the thermal decomposition processes of 1-3 were investigated by simultaneous TG-DSC techniques. The TG curves reveal that complexes 1-3 possess pretty good thermostability up to 321 degrees C, 416 degrees C and 418 degrees C, respectively. The apparent activation energy E (E-1 = 194.975 kJ mol(-1), E-2 = 220.360 kJ mol(-1), E-3 = 223.600 kJ mol(-1)) and the pre exponential factor A of skeleton collapse for the complexes 1-3 were calculated by the integral Kissinger’s method and Ozawa-Doyle’s method. The structural stability could be illustrated from the point of thermal decomposition kinetics.

Insulin resistance is thought to be a common link between obesity and Non-Alcoholic Fatty Liver Disease (NAFLD). NAFLD has now reached epidemic status worldwide and identification of molecules or pathways as newer therapeutic strategies either to prevent or overcome insulin resistance seems critical. Dysregulated hepatic lipogenesis (DNL) is a hallmark of NAFLD in humans and rodents. Therefore, reducing DNL accretion may be critical in the development of therapeutics of NAFLD. In our in vivo model (high-fat-diet fed [HFD] obese mice) we found Zinc oxide nanoparticles (ZnO NPs) significantly decreased HFD-induced hepatic steatosis and peripheral insulin resistance. This protective mechanism of ZnO NPs was signaled through hepatic SIRT1-LKB1-AMPK which restricted SREBP-lc within the cytosol limiting its transcriptional ability and thereby ameliorating HFD mediated DNL. These observations indicate that ZnO NP can serve as a therapeutic strategy to improve the physiological homeostasis during obesity and its associated metabolic abnormalities. (C) 2019 Elsevier Inc. A series of novel 2-(5-aminomethylene-4-oxo-2-thioxothiazolidin-3-yl)-3-phenylpropionic acid ethyl esters has been synthesized. Target compounds were evaluated for their trypanocidal activity towards Trypanosoma brucei brucei and Trypanosoma brucei gambiense. Several hit-compounds (8, 10, 12) inhibited growth of the parasites at sub-micromolar concentrations (IC50 0.027-1.936 mu M) and showed significant selectivity indices (SI= 108-1396.2) being non-toxic towards the human primary fibroblasts. The screening of anticancer activity in vitro within NCI DTP protocol allowed to identify active 2-(5-{[5-(2,4-dichlorobenzyl)-thiazol-2-ylamino]methylene}- 4-oxo-2-thioxothiazolidin-3-yl)-3-phenylpropionic acid ethyl ester 14 that demonstrated inhibition against all 59 human tumor cell lines with the average GI(50) value of 2.57 mu M. It was established that the activity type (antitrypanosomal or anticancer) as well as its level depends on the character of enamine fragment in the C5 position of thiazolidinone core.

A totally new theory of two-step stabilization mechanism for triazole-based zinc-containing complex (abbreviated as (Zn(ttr)(OAc)]) was proposed through several designed experiments. [Zn(ttr)(OAc)] strongly absorbed HCl at the beginning, and then broke down into 3-amino-1,2,4-triazole which could further stabilized PVC. The structure of [Zn(ttr)(OAc)] was characterized by FTIR spectroscopy, elemental analysis and SEM. Then, Congo red tests and discoloration tests verified [Zn(ttr)(OAc)] could absorb HCl effectively, but PVC showed poor initial whiteness, which further proved above -mentioned stabilization mechanism. Moreover, [Zn(ttr)(OAc)] combined with ZnSt(2) and dibenzoyl methane (DBM) effectively improved the initial color of PVC samples and provided PVC with shorter plasticizing time and longer dynamic thermal stability time than commercial stabilizer 1086F, confirmed by discoloration tests and dynamic thermal stability analysis. We believe [Zn(ttr)(OAc)]/ZnSt(2)/DBM stabilizer system can be used as a commercial stabilizer and the stabilization mechanism of [Zn(ttr)(OAc)] may provide a new way to stabilize PVC. A highly sensitive dual-signal aptamer sensor based on polydopamine-gold nanoparticles (PDA-AuNPs) and exonuclease I (Exo I) was developed for detection of malathion. Compared with traditional sensing elements, aptamer has many advantages, such as high affinity, superior specificity, strong stability and easy modification. The electrodeposition synthesis of PDA-AuNPs gave excellent biocompatibility and electrical conductivity on the sensor. With the addition of malathion, the specific interaction between malathion and its aptamer forced the aptamer to detach from the electrode surface and induced the capture probe to form a hairpin structure on the electrode surface. Exo I was added to motivate the autocatalytic target cycling which remarkably increased the current change of electrochemical signal over 2 times. Therefore, the promising strategy gave rise to an optional dual-signal current readout in both the signal-on of Fc and the signal-off of Tn. In this work, the prepared biosensor exhibited high sensitivity to malathion via the combination of dual-signal design and autocatalytic target cycling amplification. Under the optimized conditions, the proposed sensor showed a wide linear range from 0.5 to 600 ng/L malathion. It also exhibited excellent specificity, acceptable repeatability and good stability. The application of real samples obtained satisfactory recovery results, demonstrating a promising potential in food safety analysis.

A novel pyridinium salt derivative, 2,3,4,5,6-pentaphenyl-1-(1H-1,2,4-triazol-3-yl)pyridinium perchlorate (1) has been designed and synthesized by using pyrylium perchlorate and 3-amino-1H-1,2,4-triazole as ingredients, which can be exploited for the binding and specific sensing of anion via hydrogen bonds. Compound 1 exhibits intense blue emission in solution with maximum emission at 469 nm with a red shift relative to that (426 nm) of solid state. The luminescence intensity of 1 could be selectively and sensitivity quenched with acetate ion and fluorine ion among various anions including F-, Cl-, Br-, I-, AcO- and NO3-. Furthermore, the possible detection mechanism was investigated. This work could contribute a credible potential sensing application of 1,2,4-triazolyl substituted pentaphenylpyridinium salts towards the specific anions in biological and environmental concerns. Cancer is a leading cause of human death worldwide. One of the greatest challenges in cancer therapy is the discovery and design of novel products with potential anti-tumor activities. In this study, a new protocol involves three-component condensation of the 3-amino-1,2,4-triazole as a 1,3-binucleophile, versatile aldehydes and Nmethyl-1-(methylthio)-2-nitroethenamine as an enamine analogous in the presence of trichloroacetic acid as a Bronsted-Lowry acidic promoter leads to new functionalized N-alkyl-6-nitro-3,5-dihydro-[1,2,4]triazolo [1,5-a] pyrimidin-7-amine in moderate to good yields. The presence of five nitrogen heteroatoms in the product structure has gathered immense attention among chemists and biologists due to their biological values. Therefore, we evaluated the anti-tumor activity of our synthetic compounds on different cancer cells including human malignant melanoma cells (A375), prostate cancer cells (PC3 cells, LNCaP cells) and normal cells HDF (human dermal fibroblast). Notably, we found that compound 4b that contains a nitro group has the best antitumor activity on three different cancer cells. By using DAPI staining, we showed cancer cells death. Apoptosis induction was shown using quantitative real time PCR (qRT-PCR) by evaluating of Bax and Bcl2 mRNA levels. Finally, we demonstrated that 4b has epithelial-to-mesenchymal transition (EMT) inhibition effect on cancer cells (by induction of E-cadherin and reduction of vimentin mRNA expression levels as two potential EMT markers). So, 4b could be an anti-cancer promising drug. Although, in vivo experiments will be required to evaluate possible side effects.

Neutrophil survival and oxidative stress during inflammatory conditions are linked to tissue damage. The present study explores less understood role of catalase, the enzyme catalysing hydrogen peroxide decomposition, in neutrophil survival/death. Importantly, inhibition of catalase activity following S-glutathionylation in the PMA, NO, or zymosan-activated neutrophils or treatment with catalase inhibitor led to neutrophil death. On the contrary, introducing reducing environment by TCEP rescued catalase activity and significantly improved neutrophil survival. Furthermore, augmentation in ROS generation by NOX-2 activation or induction of mitochondrial ROS by Antimycin-A induced catalase S-glutathionylation and cell death, which was prevented in the neutrophil cytosolic factor1 (NCF-1-/-) cells or was rescued by MitoTEMPO, a mitochondrial ROS scavenger, thus, suggesting a correlation between catalase S-glutathionylation/activity inhibition and reduced neutrophil survival. Altogether, enhanced NOX2 activation/mitochondrial dysfunction led to reduced survival of human and mice neutrophils, due to H2O2 accumulation, S-glutathionylation of catalase, and reduction in its enzymatic activity. The present study thus demonstrated mitigation of catalase activity under oxidative stress-impacted neutrophil survival.

The reaction of N-(4-acetylphenyl)benzene sulphonamide derivatives 3a and 3b with N,N dimethyl formamide dimethyl acetal (DMF-DMA) afford acryloyl(phenyl)benzenesulfonamide derivatives 4a and 4b; respectively. The chemical reactivity of enaminonitriles 4a and 4b towards hydrazine hydrate or hydroxylamine was studied for synthesizing of pyrazolyl and isoxazolyl-phenyl benzenesulfonamide derivatives 5a,b and 6a,b; respectively. Also, the treatment of enaminonitriles 4a and 4b with thiosemicarbazide or heterocyclic amines derivatives afford the novel sulfonamide derivatives. Furthermore, the reactivity of acetylsulfonamide derivatives towards nitrogen nucleophiles and dimedone afforded novel benzene sulfonamide compounds. Some of the synthesized chlorinated compounds exhibited excellent in vitro antitumor activity against HepG2 and MCF-7 cell lines. Additionally, further studies were carried out on one of the most effective compounds, 4-chloro-N-(4-(isoxazole-3-yl)phenyl) benzenesulfonamide 6a (ISP), to evaluate its potential interaction against KSHV thymidylate synthase complex. The comprehensive theoretical and experimental mechanical studies of compound 6a (ISP) were compatible with elemental analysis, FTIR, H-1 NMR and MS spectral data. The optimized molecular structure and the harmonic vibrational frequencies were examined via Density functional theory (DFT)/B3LYP/6-31G(d) and Hartree-Fock HF/6-31G(d) energies.
