A facile strategy for the synthesis of steroidal [1,2,4]triazolo[1,5-a]pyrimidines 5a-g and 6a-g has been accomplished via a one-pot reaction of steroidal ketones, aromatic aldehydes and 3-amino-1,2,4-triazole in the presence of potassium tert-butoxide in refluxing tert-butanol. All the synthesized heterosteroids were evaluated for in vitro antiproliferative activity against human cancer cells by sulforhodamine B (SRB) assays. Water is essential for honey bees (Apis mellifera L.), but contaminated sources of water in agricultural environments represent a risk of exposure to potentially harmful contaminants. Providing clean water to honey bees could be an efficient and cost-effective measure for beekeepers to reduce bee mortality associated with pesticides and improve the health of their colonies. The main goal of this study was to design a waterer prototype to fulfill the water requirements of honey bees and to evaluate the potential of this waterer in improving colonies’ health in agricultural settings, through mitigating the possible impact of an exposure to pesticides from puddle water. We tested the preference of honey bees regarding water composition and waterer prototypes, among which honey bees showed a strong preference for salted water and a poultry-type waterer. Our waterer models were quickly adopted and intensively used through the season in both the context of honey production in field crops and pollination services in cranberry crops. However, in neither context did the use of waterers reduce worker mortality nor increase overall colony weight. Our waterers provided bees with water containing fewer pesticides and were associated with reduced risks of drowning compared to natural sources of water. Our study suggests that the use of waterers fulfills an important requirement for honey bees and represents an interesting and convenient precautionary measure for beekeepers.

Key message NCP1, a NINJA family protein lacking EAR motif, acts as a negative regulator of ABA signaling by interacting with and inhibiting the activity of transcriptional activator ABP9. The phytohormone abscisic acid plays a pivotal role in regulating plant responses to a variety of abiotic stresses including drought and salinity. Maize ABP9 is an ABRE-binding bZIP transcription activator that enhances plant tolerance to multiple stresses by positively regulating ABA signaling, but the molecular mechanism by which ABP9 is regulated in mediating ABA responses remains unknown. Here, we report the identification of an ABP9-interacting protein, named ABP Nine Complex Protein 1 (NCP1) and its functional characterization. NCP1 belongs to the recently identified NINJA family proteins, but lacks the conserved EAR motif, which is a hallmark of this class of transcriptional repressors. In vitro and in vivo assays confirmed that NCP1 physically interacts with ABP9 and that they are co-localized in the nucleus. In addition, NCP1 and ABP9 are similarly induced with similar patterns by ABA treatment and osmotic stress. Interestingly, NCP1 over-expressing Arabidopsis plants exhibited a reduced sensitivity to ABA and decreased drought tolerance. Transient assay in maize protoplasts showed that NCP1 inhibits the activity of ABP9 in activating ABRE-mediated reporter gene expression, a notion further supported by genetic analysis of drought and ABA responses in the transgenic plants over-expressing both ABP9 and NCP1. These data together suggest that NCP1 is a novel negative regulator of ABA signaling via interacting with and inhibiting the activity of ABP9.

1-(1,2,4-Triazol-3-yl)-1H-tetrazole (trt) and 1-(1-tert-butyl-1,2,4-triazol-3-yl)-1H-tetrazole (btrt) were synthesized by heterocyclization of corresponding amines with triethyl orthoformate and sodium azide. The interaction of trt with copper(II) chloride in ethanol in the presence of hydrochloric acid was found to result in 10 coordination polymer [CuCl2(trt)(H2O)](n)center dot nH(2)O. N-tert-butylated ligand btrt reacts with the copper salt in ethanol giving 2D coordination polymer [CuCl2(btrt)](n). Tetrazole ring-opening reaction with the formation of [CuCl2(3-amino-1,2,4-triazolium)(2)][CuCl4]center dot 2H(2)O was observed in the interaction of trt with copper(II) chloride in aqueous hydrochloric acid. The temperature-dependent magnetic susceptibility measurements of [CuCl2(btrt)](n) revealed that the copper(II) ions were weakly ferromagnetically coupled showing a coupling constant J of 1.2 cm(-1). Novel linear cationic poly(amide aminotriazole)s (PATnD) with secondary amine groups in the backbone were obtained by using azide-alkyne 1,3-dipolar cycloaddition reactions: metal- and solvent-free (thermal conditions, PATTnD) or copper(I)-catalyzed (Sharpless conditions, PATCnD). PATnD were investigated in vitro against strains of E. coli, P. aeruginosa, S. aureus, and S. epidermidis. Hemolytic activity was tested using human red blood cells (hRBC), and very low or no hemolytic activity was observed. The cytotoxicity of PATnD polymers against Human Gingival Fibroblasts (HGnF) cells was concentration-dependent, and significant differences between PATT1D and PATC1D were observed. The ability of these polymers to induce resistance against both Grampositive and Gram-negative bacteria was also assessed. Studied bacterial strains acquired resistance to catalytic polymers (PATCnD) in initial passages meanwhile resistance to thermal polymers (PATTnD) appears in later passages, being the increase of the minimum inhibitory concentration lower than in catalytic polymers. This result, together with the higher biocidal capacity of thermal polymers compared to catalytic ones, seems to suggest an influence of the regiospecificity of the polymers on their antibacterial characteristics. This study also demonstrates that PAT1D polymers, which do not appear to have strong hydrophobic residues, can exert significant antimicrobial activity against Gram-positive bacteria such as S. epidermidis. This pair of polymers, PATC1D and PATT1D, displays the greatest antimicrobial activity while not causing significant hemolysis along with the lowest susceptibility for resistance development of the polymers evaluated.

In the present investigation, some novel nitro Mannich bases derived from Michael type addition of activated nitro olefin, beta-nitrostyrene with various amines either primary or secondary including some amino sugars were designed and synthesized. The produced Mannich bases have been full characterized through different spectroscopic techniques. Antimicrobial evaluation has been performed against the Gram positive S. aureus and methicillin-resistant S. aurues (MRSA) infections. 5 of the synthesized compounds represent the best candidates in the biological screening, they have exhibited good activity with MIC values range from 100 to 250 mu g/ml. The active agents have been tested for pyruvate kinase inhibition activity with % of inhibition range from 30 to 79 % with IC50 in a nano molar range. They also exhibited significant Pyruvate kinase inhibition in nanomolar range with IC50 of 1066, 662, 1887, 418 and 1128 ng/ml, respectively (versus 196 ng/ml for AZD7545). Molecular docking calculations for active agents were performed. A complete conformational analysis molecular modeling utilizing Gaussian 09 program (HF/DFT) was used to verify the mode of bonding through the optimized geometries as well as essential quantum parameters were calculated using frontier energies (EHOMO & ELUMO) for the active candidates indicating the overall stability of the structure.

Breast cancer risk from pesticides may be missed if effects on mammary gland are not assessed in toxicology studies required for registration. Using US EPA’s registration documents, we identified pesticides that cause mammary tumors or alter development, and evaluated how those findings were considered in risk assessment. Of 28 pesticides that produced mammary tumors, EPA’s risk assessment acknowledges those tumors for nine and dismisses the remaining cases. For five pesticides that alter mammary gland development, the implications for lactation and cancer risk are not assessed. Many of the mammary-active pesticides activate pathways related to endocrine disruption: altering steroid synthesis in H295R cells, activating nuclear receptors, or affecting xenobiotic metabolizing enzymes. Clearer guidelines based on breast cancer biology would strengthen assessment of mammary gland effects, including sensitive histology and hormone measures. Potential cancer risks from several common pesticides should be re-evaluated, including: malathion, triclopyr, atrazine, propylene oxide, and 3-iodo-2-propynyl butylcarbamate (IPBC).

This work contributes to the growing interest in predictions linked with energetic salts. A reliable method to accurately compute the heat of formation of triazole-based salts was investigated. Calculations were based on Born-Haber energy cycles: gas-phase enthalpy of ions and lattice enthalpy were calculated separately. Ten triazole-based salts were synthesized and fully characterized. Their heat of combustion was measured by bomb calorimeter. Gas-phase heat of formation of cations and anions were computed at four different levels of theory: B3LYP 6-31G(d,p), CBS-4M, CBS-QB3, and G4. Ionic volumes were calculated at the B3LYP 6-31G(d,p) level with and without corrections. Lattice enthalpy estimations, based on calculated ionic volumes, were performed with the help of Jenkins and Gutowski methods. Combinations of the obtained results (gas-phase enthalpy of ions and lattice enthalpy) were used in the Born-Haber approach to predict solid phase enthalpy of formation of studied energetic salts. Direct comparison with experimental measurements enabled the identification of the most reliable path for energetic salt standard enthalpy of formation prediction.

In this work, the coordination-based energy sacrificial bonds have been constructed in the interphase between lignin and polyolefin elastomer to prepare high performance lignin-based thermoplastic elastomers (TPEs). The strength and toughness of lignin-based TPEs can be adjusted by choosing different nitrogen heterocyclic compounds as reactive assistants and Fe3+ or Zr2+ as metal coordination centers. It was demonstrated that 3-Amino-1,2,4-triazole with three nitrogen atoms in the heterocydic ring and one nitrogen branch chain could form the most efficient coordination bond system and generate the best mechanical performance. The system with ferric iron as coordination center exhibited better enhancement effect than divalent zinc. By adjusting the nitrogen-containing reactive additives or metal salts as coordination centers, the mechanical performance of the lignin-based TPE can be regulated, which provides a method for making green bio-composites with good strength and toughness, and also promotes the high value utilization of lignin in polymer materials.
