The title compound was prepared according to the patent application WO2008/135826. To a 50 mL 2-necked flask containing 1-methyl-1H-1,2,3-triazole (1.0 g, 12.0 mmol, prepared according to PCT Int. Appl., 2008098104) was added THF (45 mL) and the colorless solution was cooled to -40 C. Then, n-BuLi (2.5 M in hexanes, 4.8 mL, 12.0 mmol) was added dropwise which afforded a dark reddish-brown viscous solution. The mixture was stirred between -30 to -20 C. for 45 minutes, then neat DMF (3 mL, 38.5 mmol) was introduced at -10 C. The mixture was allowed to warm up to room temperature and stirred for 60 minutes, followed by pouring into water. The aqueous portion was extracted with EtOAc (4*50 mL) and the combined organics were washed with brine, dried over MgSO4, filtered and concentrated. The aqueous portion was back-extracted with DCM (3*50 mL) and dried as above. The combined organics were concentrated to give a light brown oil that was much more UV active than the starting material. TLC in either 25% CH3CN-DCM or 25% EtOAc-DCM showed the product to have a slightly higher Rf than the starting material. Chromatography on silica gel (100% DCM increasing to 25% CH3CN-DCM) provided the title compound as a colorless oil.

The cooled mixture was adjusled to pHi2 with 1M sodium hydroxide and extracled with EtOAc (50 mL x 3). The aqueous layer was adjusted to pH 4 with I M FIC and extractedwith EtOAc (50 mLx4). The extracts was dried over Na2SO4, filtered, the filtrate concentrated in vacuo and the residue purified by chromatography on silica (Petroleum ether : EtOAc =10:1) to provide the title compound. LRMS m/z (M-f-1-1) 196.0 found, 196.0 required.

A solution of 2-iodo-5-methylbenzoic acid (4.0 g, 15.3 mmol) in DMF (10 mL) was treated with 1,2,3-triazole (2.1 g, 30.5 mmol), CsCO3 (9.95 g, 30.5 mmol), CuI (0.145 g, 0.76 mmol) and trans-N,N’-dimethylcyclohexane-1,2-diamine (0.43 g, 3.05 mmol). The mixture was heated at 120 C. for 10 min in a microwave reactor. The reaction was cooled to room temperature, diluted with water, and washed with EtOAc. The aqueous phase was acidified with 1N HCl and extracted with EtOAc. The organic layer was dried over Na2SO4, filtered and concentrated. The residue was purified by gradient elution on SiO2 (MeOH in DCM with 0.1% AcOH) to give the faster eluting 2-(2H-1,2,3-triazol-2-yl)-5-methylbenzoic acid A-2, followed by the undesired regioisomer isomer, 2-(1H-1,2,3-triazol-2-yl)-5-methylbenzoic acid. Data for A-2: 1HNMR (500 MHz, DMSO-d6) delta 12.98 (br s, 1H), 8.04 (s, 2H), 7.72-7.45 (m, 3H), 2.41 (s, 3H) ppm.

Prepared according to Mastalerz et al. J. Med. Chem. 1988, 31, 1190. To a solution of compounds A/B, step 1 (1.60 g, 3.44 mmol) in ACN:water (2:1, 45 mL) was added K2S2O8 (1.86 g, 6.88 mmol) followed by K2HPO4 (1.50 g, 8.60 mmol). The resulting mixture was heated to 90 C. for 4 h, whereupon it was cooled to rt then diluted with EtOAc and the layers separated. The aqueous layer was extracted with EtOAc (3 x) and the combined organic layers were dried over Na2SO4 and concentrated in vacuo. The crude residue was purified via silica gel chromatography (MeOH-DCM, 10%), to afford the title compounds (mixture, 670 mg, 62%) as a white solid. The mixture was further purified via chiral HPLC (Chiralcel-OJ, 2¡Á25 cm, EtOH-Hexanes, 18%), affording compound A (250 mg) and compound B (240 mg).
