Then, n-BuLi, (2.5 M in hexanes, 0.7 mL, 1.75 mmol) was added dropwise. The reaction mixture was stirred at -40 C. for 30 minutes, then tert-butyl-3-(4-chloro-2-methoxy-3-(4-(trifluoromethyl)benzyl)quinoline-6-carbonyl)-azetidine-1-carboxylate (430 mg, 0.8 mmol, Intermediate 61: step b) in 2 mL THF was introduced. The reaction mixture was allowed to warm to room temperature over 30 minutes, and was quenched after 45 minutes with aqueous NH4Cl solution. The aqueous portion was extracted with EtOAc (3*30 mL). The combined organics were washed with brine, dried over MgSO4, filtered and concentrated. Chromatography on Silica gel (10% acetone-hexane increasing to 30% acetone) afforded the title compound as a white amorphous solid. 1H NMR (500 MHz, CDCl3) delta 8.24 (d, J=2.1 Hz, 1H), 7.78 (d, J=8.8 Hz, 1H), 7.56-7.47 (m, 3H), 7.40 (d, J=8.1 Hz, 2H), 7.35 (dd, J=8.7, 2.1 Hz, 1H), 4.35 (s, 2H), 4.20 (t, J=8.8 Hz, 1H), 4.07 (s, 3H), 4.00 (dd, J=9.3, 5.6 Hz, 1H), 3.92 (dd, J=8.9, 5.7 Hz, 1H), 3.67 (s, 3H), 3.62 (t, J=8.8 Hz, 1H), 3.52-3.38 (m, 1H), 1.38 (s, 9H); MS (ESI): mass calcd: Chemical Formula: C30H31ClF3N5O4; Exact Mass: 617.2. m/z found 617.8 [M+H]+.

The combined organic extracts were sequentially washed with water (500 mL) and brine solution (100 mL). The organic layer was dried over sodium sulfate, filtered, and concentrated in vacuo. The crude reaction mixture was purified using silica gel chromatography (30-80% EtOAc/hexane, linear gradient) to afford a 1.7:1.0 mixture of two regioisomeric products as an orange solid. The orange solid was recrystallized from EtOAc (hot- cold) to provide 4.25 g of l-(2,6-difluoro-4-nitrophenyl)-3-melhyl-lH-l.,2,4-triazole as a white solid. The mother liquor was recrystalized from EtO Ac/Hex (hot-rt) to afford 1.89 g of l-(2,6-difluoro-4-nitrophenyl)-5-methyl-lH-l,2,4-triazole as a light yellow solid. The mother liquor was repeatly chromatographed under the previously stated conditions to provide two samples. One sample was partially enriched in the slightly less polar isomer, l-(2,6-difluoro-4~nitrophenyl)-3-memyl-lH-l,2,4-t?azole. The other sample was partially enriched in the slightly more polar isomer, l-(2,6- difluoro-4-nitrophenyl)-5-methyl-lH-l,2,4~triazoe. Both samples were independently recrystallized from EtO Ac/Hex to provide an additional 1.9 g of 1- (2,6-diiluoro-4-nitrophenyl)-3 -methyl- 1 H- 1 ,2,4-triazole and 1.37 g of l-(2,6- difluoro-4-nitrophenyl)-5-methyl- 1 H- 1 ,2,4-triazole. A combined total of 6.15 g (26 % yield) of 1 -(2,-difluoro-4-nitrophenyl)~3-methyl-l H- 1 ,2,4-triazole and 3.2 g (13% yield) of l-(2,6-difluoro-4-nitrophenyl)-5-methyl- IH-1 ,2,4-triazole was obtained.Data for 1 -(2 s6-difluoro-4-nitrophenyl)-3 -methyl- 1 H- 1 ,2,4-triazole: LC-MS(M+H)+ = 241.0. 1H NMR (500 MHz, CHLOROFORM-d) delta ppm 8.34 (s, 1 H), 8.01- 8.10 (m, 2 H)5 2.54 (s, 3 H). Data for l-(2,6-difluoro-4-nitrophenyl)-5-methyl-lH-l,2,4-triazole: LC-MS(M+H)+ = 241.0. IH NMR (500 MHz, CHLOROFORM-d) delta ppm 8.08 (s, 1 H)5 8.04- 8.08 (m, 2 H), 2.45 (s, 3 H). An X-ray analysis of a single crystal grown from EtOAc/hexanes confirmed the regiochemical assignment of this isomer as the 5- methyl congener.

LC-MS m/z 217.0 (M+H)+.

The solution was stirred at 80 C for 2.5 h. After the mixture was cooled to room temperature, water and EtOAc were added and the phases were separated. The organic phase was washed with water, brine, dried (Na2S04) and the solvent was removed in vacuo. The residue was purified by Biotage silica gel column chromatography eluting with DCM/EtOAc (99/1 to 90/10, 10 g column) to afford the title product as a pale yellow solid (82 mg, 70.7 %). 1 H NMR (500 MHz, CDCI3): <5 4.04 (s, 3H), 4.14 (s, 3H), 7.10-7.13 (m, 2H), 7.82 (s, 1 H), 7.98-8.01 (m, 1 H); LC (Method B)-MS (ESI, m/z) fR 1 .97 min, 235 [(M+H+), 100%]. It has been reported that the salts of 1,2,4-triazole and 3-amino-1,2,4-triazole could be obtained via an acid-base neutralization [29, 30]. Likewise, the LiDAtz with two amino groups on the triazole ring was prepared by the neutralization from equimolar 3,5-diamino-1H-1,2,4-triazole (DAtzH) and LiOH. Briefly, equimolar DAtzH and LiOH were added into deionized water separately, mixed and kept reacting and stirring at room temperature for 24 h, and the synthetic process is listed in Scheme 1. The product, a yellow powder, was obtained with 96.8% yield after evaporation of water and then drying at 120 C under vacuum for 24 h. 13C Nuclear magnetic resonance (NMR, 400 MHz, D2O): 161.73 ppm. Fourier transform infrared (FT-IR, KBr, cm-1) spectra: 3374, 3417 (w, nuNH2), 3335 (m, nuH2O), 1608 (m, deltaNH2), 1558 (m, nuC-N), 1475 (s, nuC-N), 1382 (m, nuC-N-C), 1070 (m, nuC-N), 688, 773 (w, deltaN-C-N). Elemental analyses (EA; C2H4N5Li, 105.06): found: C 13.22, H 2.20, N 78.42%; calculated: C 13.20, H 2.22, N 76.96%. Combined toluene layer is washed with water ( 1 3000ml), washed toluene layer is dried over anhydrous sodium sulphate, filtered and evaporated toluene to obtain crude anastrozole (45Og) having isomeric impurity 0.5%. EXAMPLE 5 Preparation of crude Anastrozole:To the title compound of example 2 (5Og) in dry DMF (250ml) anhydrous potassium carbonate (26g) and triazole sodium salt (232 g) are added and the mixture stirred at room temperature for 45 min. Worked up the reaction mixture by adding water (750 ml) and extracting with toluene (3 x 300ml). Combined toluene layer is washed with water ( 1 x300ml).The washed toluene layer is dried over anhydrous sodium sulphate, filtered and evaporated toluene to obtain crude anastrozole (42g) having isomeric impurity 4.52%.EXAMPLE 6Preparation of crude Anastrozole: To the title compound of example 2 (5Og) in dry DMF (250ml) anhydrous potassium carbonate (26g) and triazole sodium salt (232 g) are added and the mixture stirred at room temperature for 45 min. Worked up the reaction mixture by adding water (950 ml) and extracting with toluene (3 x 300ml). Combined toluene layer is washed with water ( 1 x30ml).The washed toluene layer is dried over anhydrous sodium sulphate, filtered and evaporated toluene to obtaincrude anastrozole (42g) having isomeric impurity 2.6%.EXAMPLE7Preparation of crude Anastrozole:To the title compound of example 2 (5Og) in dry DMF (250ml) anhydrous potassium carbonate (26g) and triazole sodium salt (232 g) are added and the mixture stirred at room temperature for 45 min. Worked up the reaction mixture by adding water (2500 ml) and extracting with toluene (3 x 300ml). Combined toluene layer is washed with water ( 1 .300ml).The washed toluene layer is dried over anhydrous sodium sulphate, filtered and evaporated toluene to obtain crude anastrozole (42g) having isomeric impurity 3.2%.

The layers were separated. The organic layer was washed with half-saturated aqueous sodium chloride solution (3 100 mL). The washed solution was dried with sodium sulfate, and the dried solution was filtered. The filtrate was concentrated to provide an off-white solid. The solid was suspended in a mixture of heptanes and isopropyl acetate (5: 1, 600 mL). The mixture was filtered and the filter cake was washed with heptanes-isopropyl acetate (5: 1). The solids were collected and dried under vacuum to afford the title compound as a white solid.

With completing the reaction, 3ml ethanol waspoured and the magnetic nanocomposite was separated in thepresence of a magnetic stirring bar; the reaction mixture becameclear. The crude product was recrystallized from ethanol to give thepure product. The pure products were characterized by conventionalspectroscopic methods. Physical and spectral data for theselected compounds are represented below.

New discoveries in chemical research and development in 2021. The transformation of simple hydrocarbons into more complex and valuable products via catalytic C–H bond functionalisation has revolutionised modern synthetic chemistry. 27808-16-8, name is 4-Methyl-1H-1,2,3-triazole belongs to triazoles compound, it is a common compound, a new synthetic route is introduced below. Formula: C3H5N3

General procedure: Methyl 2,6-difluoro-4-{[(4-iodophenyl)sulfonyl]amino}benzoate (4.5 g, 9.9 mmol) was dissolved in N-methylpyrrolidone (90 ml), and 1,2,3-triazole (1.50 g, 21.7 mmol), copper(I) iodide (1.20 g, 6.30 mmol), potassium phosphate (6.0 g, 28 mmol), and trans-N,N?-dimethylcyclohexane-1,2-diamine (1.50 g, 10.5 mmol) were added thereto, followed by stirring at 145 C. for 12 hours in the presence of nitrogen gas. After cooling to room temperature, the reaction solution was diluted with water, followed by extraction with ethyl acetate (150 ml×5). The extraction liquids were combined, washed with saturated aqueous sodium chloride, and dried over anhydrous sodium sulfate. Then, the solvent was removed. The obtained residue was purified by silica gel column chromatography (petroleum ether/ethyl acetate=2:1 to 1:1) to obtain methyl 2,6-difluoro-4-({[4-(1H-1,2,3-triazol-1-yl)phenyl]sulfonyl}amino)benzoate (M-17) (650 mg, 17%) and methyl 2,6-difluoro-4-({[4-(2H-1,2,3-triazol-2-yl)phenyl]sulfonyl}amino)benzoate (M-18) (800 mg, 20%). The title compound (252 mg, 4%) was obtained as a white solid by subjecting methyl 2,5-difluoro-4-[(4-iodophenyl)sulfonylamino]benzoate (7.00 g, 15.5 mmol) and 4-methyl-1H-triazole (2.50 g, 30.1 mmol) to the same methods as in Examples 15 to 18. [1338] 1H NMR (400 MHz, DMSO-d6): delta 11.18 (s, 1H), 8.67 (s, 1H), 8.15 (m, 2H), 8.06 (m, 2H), 7.68-7.63 (m, 1H), 7.34-7.29 (m, 1H), 3.80 (s, 3H), 2.33 (s, 3H)
