Targeting bromodomain-containing protein 4 (BRD4) has been proved to be an effective strategy for cancer therapy. To date, numerous BRD4 inhibitors and degraders have been identified, some of which have advanced into clinical trials. In this work, a focused library of new [1,2,4]triazolo[1,5-a]pyrimidine derivatives were discovered to be able to inhibit BRD4. WS-722 inactivated BRD4 (BD1/BD2), BRD2 (BD1 BD2) and BRD3 (BD1/BD2) broadly with the IC50 values less than 5 mu mol/L. Besides, WS-722 inhibited growth of THP-1 cells with an IC50 value of 3.86 mu mol/L. Like (+)-JQ1, WS-722 inhibited BRD4 in a reversible manner and enhanced protein stability. Docking studies showed that WS-722 occupied the central acetyl-lysine (Kac) binding cavity and formed a hydrogen bond with Asn140. In THP-1 cells, WS-722 showed target engagement to BRD4. Cellular effects of WS-722 on THP-1 cells were also examined, showing that WS-722 could block c-MYC expression, induce G0/G1 phase arrest and p21 up-regulation, and promote differentiation of THP-1 cells. BRD4 inhibition by WS-722 resulted in cell apoptosis and up-regulated expression of cleaved caspased-3/7 and PARP in THP-1 cell lines. The [1,2,4]triazolo[1,5-a] pyrimidine is a new template for the development of new BRD4 inhibitors. Co-crystals are emerging as members of supramolecular family having particular practical and fundamental significance for chemists, crystallographers, pharmaceutical scientists, and theoreticians. The presented study is focused to synthesize anti-cancer co-crystals of commercially available nandrolone (1), a synthetic anabolic-androgenic steroidal drug. Co-crystallization is done using economical and green grinding and reflux methods to obtain nandrolone (Nan): salicylic acid (Sal) and nandrolone (Nan): 3-amino-1,2,4-triazole (Triz) co-crystals, in 2:1 and 1:1 stoichiometric ratios, respectively. The structural analysis and characterization were carried out using single-crystal X-ray diffraction, and vibrational spectroscopy. Nandrolone (1) crystallizes in monoclinic P2(1) space group, while the co-crystal-I (Nan:Sal) and co-crystal-II (Nan:Triz), co-crystallized in the orthorhombic P2(1)2(1)2(1) space group. The intermolecular hydrogen bonds O-H center dot center dot center dot O, C-H center dot center dot center dot O, N-H center dot center dot center dot O, O-H center dot center dot center dot N, and N-H center dot center dot center dot N between the active pharmaceutical ingredient (nandrolone) and co-formers (salicylic acid, and 3-amino-1,2,4-triazole) stabilize the structures of cocrystals. In vibrational spectroscopy of co-crystal-I (Nan:Sal), the blue shifts in stretching frequencies of hydroxyl group from 3417.9 cm(-1) to 3427.8 cm(-1) further supported the hydrogen bond interactions between API and co-former. Similarly, in co-crystal-II (Nan:Triz) the NH2 stretching frequency from 3331.4- 3413.4 cm(-1) to 3312.7 cm(-1), supported the interaction of NH2 with API via intermolecular interaction. Nandrolone (1) and both co-crystals were found to be non-cytotoxic against 3T3 normal fibroblast cell line. Nandrolone (IC50 = 1.0 0.1 mu M), co-crystal-I (Nan:Sal) (IC50 = 1.6 +/- 0.3 mu M) and-II (Nan:Triz) (IC50 = 1.8 +/- 0.1 mu M) showed anti-cancer potential against cervical cancer HeLa cell line. While doxorubicin (IC50 = 1.2 +/- 0.2 mu M) was used as standard tested compound. SYNOPSIS Two new non-cytotoxic co-crystals of synthetic anabolic-androgenic steroidal drug nandrolone (Nan) with pharmaceutically acceptable salicylic acid (Sal), and triazole (Triz) were synthesized and their structures were elucidated using single-crystal X-ray diffraction, and vibrational spectroscopy. Quantitative analysis of -OH and -NH2 intermolecular interactions between API and co-former by Hirshfeld surface analysis further supported the role of various functionalities towards the stability of co-crystals. Both co-crystals were found to be non-cytotoxic against 3T3 normal fibroblast cell line. Co-crystal-I (Nan:Sal) and co-crystal-II (Nan:Triz) were found to be selectively active against HeLa cancer cell line (IC50 = 1.6 +/- 0.3 mu M), (IC50 = 1.8 +/- 0.1 mu M). (c) 2020 Elsevier B.V. In the present study, the chemical structure of 4,4′-trimethylene-N,N’-dipiperidinium chlorosulfonate is elucidated by 1D NMR, and 2D NMR, FTIR, Raman analyses, and Mass spectra. It was indicated that the chlorosulfonic acid could not act as a sulfonating agent in the reaction with secondary amines like TMDP due to due more steric hindrance of the sulfur atom by chlorine and oxygen atoms. Raman and FTIR spectra are demonstrated as two high-value optical techniques to the elucidation and identification of chemical structures that confirmed C3(v) symmetry for chlorosulfonate anion in the ionic liquid. Then, the solvent-catalyst activity of ionic liquid was demonstrated in the one-pot synthesis of a series of novel triazolo-pyrimidines under mild conditions. Gaseous atmosphere during graphitic carbon nitride (g-C3N4) preparation can have a significant influence on modifying the morphological texture, polymeric structure, charge carrier behavior, and consequently the photocatalytic performance. Herein, we developed a new one-step method to fabricate g-C3N4 through direct pyrolysis of 3-amino-1,2,4-triazole in CO2 atmosphere (C3N4-T-CO2) with no additive. Surprisingly, the H-2 production activity of C3N4-T-CO2 photocatalyst under visible light irradiation was 2.4 and 1.7 times as high as that of g-C3N4 obtained under air and N-2 atmosphere with the same heating process, respectively. Detailed characterizations indicated that the CO2 calcination atmosphere induced less nitrogen vacancies with no charge transport ability, more NHx groups, and faster rate of the electron transport between heptazine rings for C3N4-TCO2 among three samples. It is also suggested that the larger number of NHx in C3N4-T-CO2 could enhance the interlayer electron transport through the hydrogen-bonding interaction between C3N4 layers. Time-resolved photoluminescence, single-particle fluorescence, and femtosecond time-resolved transient absorption measurements were performed to elucidate the efficient charge transfer and trapping processes in C3N4-T-CO2. For the first time, such unprecedented effect of CO2 calcination atmosphere was observed for g-C3N4. This work not only presents a promising strategy in designing highly effective g-C3N4 photocatalyst for solar energy conversion, but also makes an insight into the charge transfer process in g-C3N4 photocatalyst.

A highly efficient multi-component one-pot synthesis of novel 1H-imidazo[1,2-a]imidazole-3-amine and imidazo[2,1-c][1,2,4]triazole-5-amine derivatives has been developed through the reaction of easily available aromatic aldehydes, benzoyl cyanide and either 2-aminoimidazole-4,5-dicarbonitrile or 3-amino-1,2,4-triazole in pyridine under controlled microwave heating. The process is environmentally friendly, is operationally simple, with short reaction time and with high yields.

A novel, eco-friendly and fast route has been developed for the synthesis of new and known triazolo[1,5-a]pyrimidin fused chromone derivatives via a one pot three-component reaction of 3-amino-1,2,4-triazoles, aromatic aldehydes and 4-hydroxycoumarin in aqueous medium at room temperature. These reactions are catalyzed by MCM-41-HWO4 as a safe and recyclable mesoporous solid acid. It combines successfully the synergistic effect of green chemistry with nanocatalysis. The yields are high and the products were characterized by H-1 NMR, (CNMR)-C-13 spectra and elemental analysis.

Bismuth oxyhalides (BiOX (X = Cl, Br, I) are considered to be an important p-type semiconductors in the photocatalysis applications. In particular, tetragonal BiOBr is considered as a stable photocatalyst due to its resilient absorption in the visible region with an band gap energy of 2.8 eV. In the meantime, lanthanide ions (with 3+ oxidation state) implies as conversion catalyst gained huge impact and remain a serious topic in materials chemistry. Here we synthesized upconversion photocatalyst mainly consists of BiOBr with the Er 3+ and Tb 3+ ions along with low band gap g-C3N5 for the improved photocatalytic performances. The synthesized Er3+/ Tb3+@BiOBr-g-C3N5 heterojunction was systematically characterized by XRD, and FT-IR for the confirmation of the composite and their morphology were analysed with FESEM and HR-TEM analysis which revealed that the sheets of g-C3N5 were decorated by Er3+/Tb3+ loaded BiOBr microspheres. The XPS analysis confirmed the suitable oxidation state of all the individual elements existing in the composite. As the UV-DRS analysis showed that the band gap of the Er3+/Tb3+ BiOBr-gC3N5 heterojunction was narrowed to 2.64 eV. To evaluate the photocatalytic efficiency of the synthesized g-C3N5, Er3+/Tb3+@BiOBr and Er3+/Tb3+@BiOBr-gC3N5 heterojunction under the simulated visible light irradiation source towards the aqueous sulfamethoxazole degradation. The Er3+/Tb3+@BiOBr-gC3N5 heterojunction shows maximum degradation efficiency of 94.2% after 60 min of visible light irradiation whereas the pure g-C3N5 provided about 43.8% and Er3+/Tb3+@BiOBr implies 55.2% degradation efficiency. The plausible degradation mechanism of pollutant removal was proposed.

Chemical transformations of chromone-3-carbonitrile (1) with some substituted hydrazines, namely, thiosemicarbazide, S-methyl/benzyldithiocarbazate, 7-chloro-4-hydrazinoquinoline, and 3-hydrazino-5,6-diphenyl-1,2,4-triazine, led to substituted pyrazoles 2, 5-8. Ring opening of carbonitrile 1 followed by recyclization with 3-amino-1,2,4-triazole and 2-aminobenzimidazole gave triazolo[1,5-a]pyrimidine 9 and pyrimido[1,2-a]benzimidazole 10, respectively. Treatment of carbonitrile 1 with some heterocyclic amines produced 2-amino-3-substituted-chromones 11 and 12. The novel 3-hydroxychromeno[4,3-b]pyrazolo[4,3-e]pyridin-5(1H)-one (13) was efficiently synthesized from the ring conversion of carbonitrile 1 with cyanoacetohydrazide. A mixture of chromeno[2,3-b]naphthyridine 14 and chromeno[4,3-b]pyridine 15 was obtained from base catalyzed transformation of carbonitrile 1 with malononitrile dimer. A diversity of novel annulated chromeno[2,3-b]pyridines 16-22 was also synthesized. Chromeno[2,3-b]pyrrole-2-carboxylate 23 was obtained from the reaction of carbonitrile 1 with ethyl chloroacetate. Structures of the new synthesized products were deduced on the basis of their analytical and spectral data.

For the first time, the coordination-based energy sacrificial bonds have been constructed in the interface between lignin and polyolefin elastomer for preparing a new class of high performance thermoplastic elastomers (TPEs) with biomass lignin as the hard plastic phase. The coordination bonds not only promoted the dispersion of lignin, but also improved the interfacial interactions between lignin and polyolefin elastomer matrix, and also facilitated the orientation of chain segments during stretching. The synergistic coordination effect of lignin promoted higher energy dissipation, leading to simultaneously enhanced strength and toughness of lignin-based TPEs even with the lignin loading as high as 30 wt %. The lignin-based TPEs also exhibited excellent shape memory performance. Our strategy offers a promising methodology for the facile production of high performance but cost-effective TPE materials using biorenewable resources as plastic phase.

Wood (secondary xylem) formation in tree species is dependent on auxin-mediated vascular cambium activity in stems. However, the complex regulatory networks underlying xylem formation remain elusive. Xylem development in Populus was characterized based on microscopic observations of stem sections in transgenic plants. Transcriptomic, quantitative real-time PCR, chromatin immunoprecipitation PCR, and electrophoretic mobility shift assay analyses were conducted to identify target genes involved in xylem development. Yeast two-hybrid, pull-down, bimolecular fluorescence complementation, and co-immunoprecipitation assays were used to validate protein-protein interactions. PaC3H17 and its target PaMYB199 were found to be predominantly expressed in the vascular cambium and developing secondary xylem in Populus stems and play opposite roles in controlling cambial cell proliferation and secondary cell wall thickening through an overlapping pathway. Further, PaC3H17 interacts with PaMYB199 to form a complex, attenuating PaMYB199-driven suppression of its xylem targets. Exogenous auxin application enhances the dual control of the PaC3H17-PaMYB199 module during cambium division, thereby promoting secondary cell wall deposition. Dual regulation of xylem formation by an auxin-mediated PaC3H17-PaMYB199 module represents a novel regulatory mechanism in Populus, increasing our understanding of the regulatory networks involved in wood formation.
