The mixture containing the 2 and 4-triazolyl-benzonitriles was purified by a combination of chromatography on silica gel (elution gradient of ethyl acetate in hexane) and on reversed phase silica gel to give 1.86 g (18% yield) of Intermediate 5 and 0.526 g (5% yield) of Intermediate 6. Intermediate 5 4-Fluoro-2-(3-methyl-1H-1,2,4-triazol-1-yl)benzonitrile: White crystals (ethyl acetate/hexanes); mp 117-118 C. 1H NMR (400 MHz, CDCl3) delta: 8.82 (1H, s), 7.84 (1H, dd, J=5.6, 8.6 Hz), 7.62 (1H, dd, J=2.5, 9.1 Hz), 7.24 (1H, m), 2.54 (3H, s). Anal. Calcd for C10H7FN4: C, 59.40; H, 3.49; N, 27.71. found: C, 59.25; H, 3.32; N, 27.81. 1-Methyl-1H-1,2,3-triazole was prepared according to the literature reference W02008/98104. To a 2 L flask containing 1-methyl-1H-1,2,3-triazole (9 g, 108.3 mmol) was added THF (1500 mL) and the solution was cooled to -40 C. To this colorless homogeneous solution was added n-butyllithium (2.5 M in hexanes, 45 mL, 112.5 mmol) dropwise which immediately afforded a dark brown viscous mixture. The mixture was kept between -10 to -20 C. for 60 minutes, then a THF solution of 2,4-dimethylthiazole-5-carbaldehyde (17.2 g, 121.8 mmol in 200 mL THF) was introduced via cannula. Once the aldehyde was added the reaction was allowed to warm to room temperature. After 3 hours, the reaction was quenched by pouring it into a saturated solution of aqueous NH4Cl. The aqueous portion was extracted with EtOAc in portions, 7*400 mL. The combined organics were washed with brine, dried over MgSO4, filtered and concentrated to afford a brown oil. Chromatography on silica gel (10% acetone-DCM increasing to 50% acetone and increasing to 10% MeOH-DCM) provided the title compound as an amber solid.

It was stirred further for 20 min at room temperature at which time LC/MS analysis confirmed full consumption of starting material. This crude compound was purification by HPLC to afford the desired title product.

The enriched material was diluted with dimethylsulfoxide and purified by reverse phase chromatography (275 gram Gold C-18 column (ISCO); 10-100percent acetonitrile/water with trifluoroacetic acid modifier). The pure fractions were concentrated to dryness under reduced pressure to give 3-bromo-1-phenyl-1,2,4- triazole (1.72 g, 38percent). 1H NMR (400 MHz, OMSO-de) delta 9.31 (s, 1H), 7.88 – 7.76 (m, 2H), 7.65 – 7.52 (m, 2H), 7.46 (t, J = 7.4 Hz, 1H) ppm. ESI-MS m/z calc. 222.9745, found 223.97 (M+l)+; Retention time: 2.66 minutes.
