Yield: 46% (610 mg, white solid). LCMS: (Method A) 174.1 (M+H), Rt.1.27 min, 67.51 % (Max).

The reaction mixture is stirred at 100 C for 2 hours. The reaction mixture is allowed to cool to room temperature, diluted with CH2C12 (100 mL), and washed with H20 (75 mL x2). The organic layer is dried with Na2S04 and filtered. The filtrate is concentrated under reduced pressure and the residue is purified by silica gel flash column chromatography with 5% MeOH in EtOAc as the eluent to afford an impure mixture of 5-chloro-2-(3-methyl- l,2,4-triazol-l-yl)-benzaldehyde plus regiosiomer (270 mg).To a mixture of 4-(2-amino-pyrimidin-5-yl)-Nl-ieri-butyl-benzene-l,2-diamine (270 mg, 1.05 mmol) and impure mixture of 5-chloro-2-(3-methyl- l,2,4-triazol-l-yl)-benzaldehyde plus regiosiomer (270 mg) in DMF (15.0 mL) is added Oxone (650 mg, 1.06 mmol) in H20 (5.0 mL). The reaction mixture is stirred at room temperature for 1.5 hours and then quenched with saturated sodium thiosulfate aq. (25 mL). The quenched reaction mixture is extracted with EtOAc (25 mL x2) and the combined organic layers are washed with H20 (50 mL x2), dried with Na2S04 and filtered. The filtrate is concentrated under reduced pressure and the residue is purified by silica gel flash column chromatography with 5% MeOH in CH2C12 as the eluent to afford an impure mixture of 5-(2-amino- pyrimidin-5-yl)- l-ieri-butyl-2-[5-chloro-2-(3-methyl- l,2,4-triazol-l-yl)-phenyl]-2,3- dihydro-lH-benzimidazol-2-ol plus regiosiomer (25 mg).

It was stirred further for 10 min at room temperature. This crude compound was purified by HPLC to afford (3-(7-(6-phenylpyridin-3-yl)-2,4-dihydro-l FIJI, 3]oxazino[5,4-e]pyrazolo[l,5-a]pyrimidin-5-yl)-8-azabicyclo[3.2. l]octan-8-yl)(4H-l?2,4- triazol-3-yl)methanone. LCMS t = 3.20 Min (10 min run, UV254nm). Mass calculated for M+ H 534.0, observed LC/MS m/z 534.0 (M+H).

The 10.0g (44.1mmol) 4,5- dibromo -2H-1,2,3- triazole (commercially available) was dissolved in 90ml of tetrahydrofuran was added 6.1g (44.2mmol) of potassium carbonate, cooled to -10 , was added 7.5g (53mmol) of methyl iodide.In the 35 ~ 40 to react until the reaction is complete.50ml of water was added, the tetrahydrofuran was distilled off, 90ml and extracted with methyl tert-butyl ether, dried over anhydrous magnesium sulfate, and concentrated to dryness under reduced pressure, the residual solid was added methyl tert-butyl ether, 10ml, 70ml of n-hexane was added slowly, solid precipitation, the addition was completed, stirring at room temperature for 1 to 2 hours.Filtered to give pure 4,5-dibromo-1-methyl -1H-1,2,3- triazole 5.8g, 57% yield.

The transformation of simple hydrocarbons into more complex and valuable products via catalytic C–H bond functionalisation has revolutionised modern synthetic chemistry. 288-36-8, name is 1H-1,2,3-Triazole, molecular formula is C2H3N3, its traditional synthetic route has been very mature, but the traditional synthetic route has various shortcomings, below Introduce a new synthetic route.

A solution of 2-iodo-5-methylbenzoic acid (4.0 g, 15.3 mmol) in DMF (10 mL) was treated with 1,2,3-triazole (2.1 g, 30.5 mmol), CsCO3 (9.95 g, 30.5 mmol), CuI (0.145 g, 0.76 mmol) and trans-N,N’-dimethylcyclohexane-l,2-diamine (0.43 g, 3.05 mmol). The mixture was heated at 120 0C for 10 min in a microwave reactor. The reaction was cooled to room temperature, diluted with water, and washed with EtOAc. The aqueous phase was acidified with IN HCl and extracted with EtOAc. The organic layer was dried over Na2SO4, filtered and concentrated. The residue was purified by gradient elution on SiO2 (0 to 10% MeOH in DCM with 0.1% AcOH) to give the faster eluting 2-(2H-l,2,3-triazol-2-yl)-5-methylbenzoic acid A-2, followed by the undesired regioisomer isomer, l-(2H-l,2,3-triazol-2-yl)-5-methylbenzoic acid. A solution of the acid (3.56 g, 17.52 mmol) in 150 mL of DCM was stirred and cooled to 00C. The solution was treated with oxalyl chloride (1.9 mL, 21.9 mmol) and DMF (68 muL, .878 mmol). The solution was slowly warmed to room temperature and stirred overnight. Solvent was concentrated and the resulting solid was azetroped with DCM and concentrated to provide A-4 as a yellow solid.

The transformation of simple hydrocarbons into more complex and valuable products via catalytic C–H bond functionalisation has revolutionised modern synthetic chemistry. 27996-86-7, name is 4-[1,2,4]Triazol-1-yl-benzaldehyde belongs to triazoles compound, it is a common compound, a new synthetic route is introduced below. Formula: C9H7N3O

L-Proline (1.81 g, 15.6 mmol) was added to a stirring, heterogeneous mixture of 4-(1H-1,2,4-triazol-1-yl)benzaldehyde (13.5 g, 78.0 mmol, Intermediate 45, step a) and 2,2-dimethyl-1,3-dioxane-4,6-dione (11.2 g, 78.0 mmol) in ethanol (520 mL) at 23 C. After 1.5 hours, diethyl 1,4-dihydro-2,6-dimethyl-3,5-pyridinedicarboxylate (19.7 g, 78.0 mmol) was added in one portion. After 16 hours, the ethanol was removed by rotary evaporation at 35 C. to afford a yellow solid. Isopropanol (300 mL) was added and the heterogeneous mixture was stirred for 10 minutes at 23 C. The mixture was filtered and the filter cake was washed with isopropanol (150 mL). The solids were collected and dried under vacuum to provide the titled compound as a white solid.
