A colorimetric and visual assay is described for the herbicide aminotriazole (ATZ). It is based on the etching of gold nanorods (AuNRs) by iodine which is formed on oxidation of iodide via H2O2. Longitudinal etching of the AuNRs occurs quickly and is accompanied by a color change from dark blue to red. In the absence of ATZ and the presence of active catalase (CAT), H2O2 is quickly decomposed into water, and the AuNRs will not be etched. In thepresence of ATZ, CAT is partially deactivated, and this affects the amount of available H2O2 and, consequently, of the iodine. Hence, the color is significantly changed. The color changes can be easily detected with bare eyes. The assay has a linear response in the 5 to 70 mu M concentration range, with a detection limit of 1.3 mu M and high selectivity for ATZ. It was applied to the determination of ATZ in water and food samples.

Insulin resistance is thought to be a common link between obesity and Non-Alcoholic Fatty Liver Disease (NAFLD). NAFLD has now reached epidemic status worldwide and identification of molecules or pathways as newer therapeutic strategies either to prevent or overcome insulin resistance seems critical. Dysregulated hepatic lipogenesis (DNL) is a hallmark of NAFLD in humans and rodents. Therefore, reducing DNL accretion may be critical in the development of therapeutics of NAFLD. In our in vivo model (high-fat-diet fed [HFD] obese mice) we found Zinc oxide nanoparticles (ZnO NPs) significantly decreased HFD-induced hepatic steatosis and peripheral insulin resistance. This protective mechanism of ZnO NPs was signaled through hepatic SIRT1-LKB1-AMPK which restricted SREBP-lc within the cytosol limiting its transcriptional ability and thereby ameliorating HFD mediated DNL. Cerrena unicolor is a wood-degrading basidiomycete with ecological and biotechnological importance. Comprehensive Biolog-based analysis was performed to assess the metabolic capabilities and sensitivity to chemicals of C. unicolor FCL139 growing in various sawdust substrates and light conditions. The metabolic preferences of the fungus towards utilization of specific substrates were shown to be correlated with the sawdust medium applied for fungus growth and the light conditions. The highest catabolic activity of C. unicolor was observed after fungus precultivation on birch and ash sawdust media. The fungus growing in the dark showed the highest metabolic activity which was indicated by capacity to utilize a broad spectrum of compounds and the decomposition of 74/95 of the carbon sources. In all the culture light conditions, p-hydroxyphenylacetic acid was the most readily metabolized compound. The greatest tolerance to chemicals was also observed during C. unicolor growth in darkness. The fungus was the most sensitive to nitrogen compounds and antibiotics, but more resistant to chelators. Comparative analysis of C. unicolor and selected wood-decay fungi from different taxonomic and ecological groups revealed average catabolic activity of the fungus. However, C. unicolor showed outstanding capabilities to catabolize salicin and arbutin. The obtained picture of C. unicolor metabolism showed that the fungus abilities to decompose woody plant material are influenced by various environmental factors.

An effectively Zn/Zn stabilizer system instead of traditional Ca/Zn stabilizer system was put forward, which contained a new triazole-based zinc-containing complex (abbreviated as [Zn(ttr)(St)]) and Zinc stearate (ZnSt(2)). Without Ca soap in the system, [Zn(ttr)(St)] itself could effectively absorb HCl and provided PVC with higher transparency and better thermal stability without zinc burning than commercial CaSt(2)/ZnSt(2) stabilizer system, verified by Congo red tests and discoloration tests and dynamic thermal stability analysis. In addition, [Zn(ttr)(St)] could further delay zinc burning of other zinc soaps, constituting Zn/Zn stabilizer system. This is attributed to the strong ability of restricting ZnCl2 of 3-amino-1,2,4-triazole released from [Zn(ttr)(St)] after absorbing HCl. After adding ESBO, PVC stabilized by [Zn(ttr)(St)]/ZnSt(2)/ESBO showed excellent transparency and thermal stability. We believe that [Zn(ttr)(St)] can act as a novel double agent with the stabilization effects of zinc soap and calcium soap at the same time and this Zn/Zn stabilizer system may inaugurate a new stabilizer system for PVC with synergistic effect. A three-descriptor quantitative structure-activity/toxicity relationship (QSAR/QSTR) model was developed for the skin permeability of a sufficiently large data set consisting of 274 compounds, by applying support vector machine (SVM) together with genetic algorithm. The optimal SVM model possesses the coefficient of determination R-2 of 0.946 and root mean square (rms) error of 0.253 for the training set of 139 compounds; and a R-2 of 0.872 and rms of 0.302 for the test set of 135 compounds. Compared with other models reported in the literature, our SVM model shows better statistical performance in a model that deals with more samples in the test set. Therefore, applying a SVM algorithm to develop a nonlinear QSAR model for skin permeability was achieved.

Five novel rhodium(II) complexes of general formula [Rh-2(mu-OOCCH3)(4)L-2], where L is a triazolopyrimidine derivative, in particular dimethyl-1,2,4-triazolo[1,5-a]pyrimidine (dmtp) for (1), 5,7-diethyl-1,2,4-triazolo[1,5a]pyrimidine (detp) for (2), 7-isobutyl-5-methyl-1,2,4-triazolo[1,5-a]pyrimidine (ibmtp) for (3), 7-hydroxy-5methyl-1,2,4-triazolo[1,5-a]pyrimidine (HmtpO) for (4) and 5,7-ditertbutyl-1,2,4-triazolo[1,5-a]pyrimidine (dbtp) for (5) are reported. These first representatives of paddle-wheel dirhodium complexes with triazolopyrimidines have been characterized by IR and NMR spectroscopy as well as by single-crystal X-ray diffraction studies. Three of the new complexes (1), (2) and (5) were thoroughly screened in vitro for their cytotoxicity against human breast cancer cell line MCF-7 and L929 murine fibroblast cells. Favorably, they show significantly less effective inhibition on the cell growth of L929 than cisplatin under identical conditions. Complexes (1) and (5) display moderate cytotoxic activity (IC50 = 16.3-21.5 mu M) against MCF-7 cells which is induced via reactive oxygen species-independent pathways. Extensive studies of rhodium complexes (1), (2) and (5) against microorganisms have shown that the tested compounds exhibit antibacterial activity against Gram-positive bacteria (Staphylococcus aureus and Bacillus subtilis) while (5) significantly inhibited the growth of Malassezia furfur. The highest antibacterial, and antifungal activity, was observed for (5).

The novel flame retarded unsaturated polyester resins have been developed and prepared by introduction of high nitrogen content additives into the polymer matrix in order to verify their effectiveness in the formation of swollen carbonaceous char inhibiting the burning process of the polymer. The intumescent flame retardants (IFRs) based on mixture or metal complex were developed and characterized by particle size distribution, Fourier-transform infrared spectroscopy (FTIR), nuclear magnetic resonance (NMR), powder X-ray diffraction (XRD), elemental analysis (CHN) and thermogravimetric analysis (TGA). The evaluation of the efficiency of IFRs addition on the flammability and smoke emission of the unsaturated polyester resins (UP) was carried out using the fire hazard (UL-94), limiting oxygen index (LOI) and cone calorimeter (CC) tests, as well as smoke density chamber tests. The volatile compounds evolved during the burning of materials were determined using a steady state tube furnace and a gas chromatograph with mass spectrometer. Furthermore, the prepared materials were subjected to differential scanning calorimetry (DSC), thermogravimetric analysis and water resistance tests. The mechanical properties of the materials were investigated using Shore D hardness and dynamic mechanical thermal analysis (DMA). The structural evaluation of the manufactured materials and samples after the cone calorimetry tests was carried out using scanning electron microscopy (SEM). It was found that the incorporation of new intumescent flame retardants led to the formation of carbonaceous char layers’ inhibiting the decomposition process and limiting the smoke emission. The most promising results were obtained for the resin containing complex designated as ZN3AT, for which the highest reduction in maximum values of heat release rate (419 kW/m(2)) compared to unmodified polymer (792 kW/m(2)) were recorded. Apart from that, the prepared intumescent flame retardants affect the cross-linking process as well as the thermal and mechanical properties of the UP.

Aminotriazole (AT), an organic heterocyclic compound, is an unselective herbicide used to deal with a large assortment of weeds by preventing the biosynthesis of carotenoids. It is a malignant agent to human beings as well as to animals. This study is focused on the nanomolar detection of AT with multiwalled carbon nanotubes along with calcium-doped zinc oxide nanoparticles adapted for a nano-composite dependent glassy carbon electrode (MWCNTs/Ca-ZnO/GCE). The developed MWCNTs/Ca-ZnO/GCE showed an excellent electrocatalytic response to AT in 3.0 pH of 0.2 M phosphate buffer solution (PBS). There was an increase in the peak current of AT, i.e., 7.726 mu A, with MWCNTs/Ca-ZnO/GCE when it was compared with the peak current of AT at the bare glassy carbon electrode (GCE), 3.478 mu A, due to the semiconducting properties of the doped nanoparticles. Various techniques were employed to study the different parameters such as scan rate, pH variations, and concentration variations by cyclic voltammetry (CV), square wave voltammetry (SWV), and linear sweep voltammetry (LSV) in order to probe the mechanism, number of participating electrons, type of process, detection limit, and quantification limit. The Ca-ZnO nanoparticles (NPs) were synthesized at the proportion of 5.0% and these were characterized by energy dispersive X-ray analysis (EDX), scanning electron microscopy (SEM), transmission electron microscopy (TEM), and X-ray diffraction (XRD). Dormancy is one of the least understood phenomena in plant biology; however, bud/corm dormancy is an important economic trait in agricultural/horticultural breeding. In this study, we isolated an ABA biosynthesis gene, GhNCED, from the transcriptome database of corm dormancy release (CDR), and characterized its negative role in regulating CDR. To understand transcriptional regulation of GhNCED, yeast one-hybrid screening was conducted and GhTCP19 was identified and shown to regulate GhNCED expression directly. An in planta assay showed that GhTCP19 negatively regulates GhNCED expression. GhTCP19 is dramatically induced by exogenous cytokinins (CKs) and is induced during CDR. Silencing of GhTCP19 in dormant cormels delayed CDR, resulting in higher expression of GhNCED and ABA levels. Meanwhile, endogenous CK biosynthesis and signaling were inhibited in GhTCP19-silenced cormels. Taken together, our results reveal that GhTCP19 is a positive regulator of the CDR process by repressing expression of an ABA biosynthesis gene (GhNCED), promoting CK biosynthesis (GhIPT) and signal transduction (GhARR) as well as inducing cyclin genes. This study expands our knowledge on CDR which is mediated by TCP family members.
