After that, ethyl methanesulfonate (2.74 g, 2.27 mL, 22 mmol) was added dropwise at room temperature. The resulting mixture was stirred for 1.5 h at 100 C, and 16 h at room temperature. After that, it was concentrated in vacuo, the residue was diluted with ethyl acetate (100 mL) and water (100 mL), the aqueous phase was extracted with ethyl acetate (3 x 100 mL). The combined organic layers were dried (sodium sulfate) and concentrated in vacuo, the residue was purified by column chromatography (silica gel, 70 g, eluting with ethyl acetate / n-heptane, gradient 0: 100 to 22:78 v/v) to yield the title compound as white solid (1.98 g, 70%). HPLC (method LCMS_fastgradient) tR = 0.88 min. 1H NMR (CDCl3, 300 MHz): delta 1.49 (t, J = 7.2 Hz, 3 H), 4.21 (q, J= 7.2 Hz, 2 H). MS (ES+) m/z 253.9, 255.9, 258.0 [M+H, 2 Br isotopes] .

General procedure: In a typical experiment Pd(OAc)2 (5.6mg, 0.025mmol), triphenylphosphine (13.2mg, 0.05mmol), iodoalkene (1-4) or iodoarene (5-19) (1mmol) were dissolved in DMF (10mL) under argon in a three-necked flask equipped with a reflux condenser and a balloon on the top. Aminotriazole nucleophile (a, b or c) (1.2mmol) and triethylamine (0.5mL) were added. The atmosphere was changed to carbon monoxide. (Caution: High pressure carbon monoxide should only be used with adequate ventilation (hood) using CO sensors as well.) The reaction was conducted for the given reaction time upon stirring at 70C. The mixture was degassed via 3 vacuum/nitrogen cycles and heated with shaking to 120 C. for 90 minutes. The reaction mixture was evaporated to dryness under reduced pressure. The residue was dissolved in water/MeOH (1:1, 3 ml) and acidified to pH=2 by addition of 2 M HCl solution. The resulting mixture was evaporated to dryness under reduced pressure then the residue was triturated with DCM/MeOH (3:1, 5 ml). The mixture was filtered washing with more DCM/MeOH (3:1, 5 ml). The filtrate was treated with trimethylsilyldiazomethane solution (2 M in hexanes, 2 ml, 4 mmol) to re-esterify the acid. The reaction mixture was evaporated under reduced pressure and the residue was purified by flash chromatography on silica gel (Biotage Snap 2×10 g columns in series, EtOAc/Cy from 50/50 to 100/0) to give the title compound D84 (48 mg) as a colourless solid. UPLC (Acid FINAL_QC_POS): rt=0.45 minutes, peak observed: 233 (M+1). C11H12N4O2 requires 232. 1H NMR (400 MHz, CDCl3) delta ppm 2.52 (s, 3H) 2.73 (s, 3H) 3.91 (s, 3H) 7.47 (d, 1H) 7.81 (d, 1H) 8.32 (s, 1H).

General procedure: Dimedone (0.14 g, 1 mmol), benzaldehyde (0.1 g, 1 mmol), and 4-phenylurazole (0.177 g, 1 mmol) were mixed with PEG-SO3H (0.6 g, 0.1 mmol) and the obtained mixture was stirred magnetically at 80 C for 40 min. After the completion of the reaction warm water (20 mL) was added and the mixture stirred for about 5 min. The insoluble crude product was filtered and recrystallized from EtOH/H2O 4:1 and the pure product was obtained (0.352 g, 91%). In order to recover the catalyst, the filtrate was dried under reduced pressure and recovered catalyst was washed with diethyl ether and reused after drying under reduced pressure. The equivalent procedure was used for the synthesis of triazolo[1,2-a]indazole-tetraones.

The aqueous fractions were further extracted with 150 mL Me-THF, then the combined organic extracts were dried (Na2SO4) and concentrated in vacuo. Purification by ISCO CombiFlash (80 g column; dry load; 100:0 to 90:10 EtOAc:EtOH over 25 minutes) provided the title compound (1-6) as a white solid. 1HNMR (400 MHz, DMSO): delta 11.46 (s, 1 H); 11.39 (s, 1 H); 7.93 (d, J=7.3 Hz, 1 H); 7.76 (s, 1 H); 7.58 (s, 1 H); 7.51 (s, 1 H); 6.67 (d, J=7.3 Hz, 1 H); 5.02 (s, 2 H).
