The resulting mixture was stirred at room temperature for 1 hour, and then concentrated in vacuo. The crude residue was purified by preparative HPLC to yield 3 products with identical masses (2 of which are diastereomers): isomer a (3.5 mg; purity 97%), isomer b (1.2 mg; purity 100%), and isomer c (1.1 mg; purity 100%). MS m/z [M+H]+ calc’d for C24H23ClFN5O33, 484.15. found 484.2.

To a mixture of l,2-dihydro-5H-l,2,4-triazol-3-one (5.74 mmol) in toluene (70 ml), al- pha,alpha-diphenylbenzenemethanol (4.7 mmol) and p-toluenesulphonic acid (1.91 mmol) were added. The reaction was heated at reflux using a Dean-Stark separa- tor under Nitrogen atmosphere for 20 hours. The solution was cooled and quenched with 2 % of an aqueous solution of NaHCO3 and extracted with CH2Cl2 (3×100 ml). The organic layer was separated, dried (Na2SO4) and the solvent evaporated. The resi- due was purified by column chromatography (eluent: CH2Cl2/Me0H 9/1). Yield: 925 mg of intermediate compound 26 (60 %).

Data for 2-triazolyl isomer: 1HNMR (500 MHz, DMSO-d6) delta 13.05 (br s , IH)5 8.12 (s, 2H), 7.81-7.52 (m, 4H) ppm. The undesired l-(2H-l,2,3-triazol-2-yl)benzoic acid eluted second. A portion of the desired acid (0.10 g, 0.53 mmol) was treated with thionyl chloride (0.59 g, 5.3 mmol) in dichloromethane (5.0 mL) at rt. After stirring at RT for 1 h, the mixture was concentrated, azeotroping with benzene to yield acid chloride A-3.

To a solution of 3,5-dibromo-1H-1,2,4-triazole (10.0 g, 44.1 mmol) in DMF (75 mL) and potassium carbonate (12.2 g, 88.2 mmol) was added iodomethane (3.02 mL, 48.5 mmol) in one portion. This gave rise to a strong exotherm from 17 C to 38 C after one minute. The reaction mixture was stirred overnight, diluted with 150 mL of EtOAc and then filtered to remove most of the inorganics. The solvent was removed under reduced pressure and the resultant yellow oily solid was partitioned between EtOAc (250 mL) and water (100 mL) and the aqueous washed with EtOAc (150 mL). The combined organics were washed with washed with brine (50 mL), dried over magnesium sulfate, filtered and the solvent removed in vacuo to give 3,5-dibromo-1-methyl-1,2,4-triazole (6.2 g, 25.8 mmol, 58% yield) as a yellow solid. UPLC-MS (ES+, Method A): 1.79 min, m/z 241.7 [M+H]+.1H NMR (400 MHz, DMSO-d6) delta 3.83 (3H).

The solvent is evaporated which affords the product R2j.1H NMR (400 MHz,DMSO-d6): delta 8.17 (s, 1 H), 4.22 (s, 3H).
