After 45 minutes, saturated aqueous ammonium chloride solution (10 mL) and ethyl acetate (100 mL) were added. The mixture was concentrated by rotary evaporation. The residue was dissolved in ethyl acetate (300 mL). The organic solution was washed with saturated aqueous sodium chloride solution (100 mL, containing excess solid sodium chloride). The aqueous layer was extracted with ethyl acetate (2*100 mL). The organic layers were combined and the combined solution was concentrated. Ether (100 mL) was added to the residue and the mixture was sonicated for 20 minutes during which time a white solid crashed out. The solids were collected by filtration. Ether (100 mL) was added to the collected solids and the mixture sonicated a second time. After 20 minutes, the mixture was filtered and the solids were collected to provide the title compound as a fine powder.

The combined organic layers were washed with water (2 x 100 mL) and brine (1 x 100 mL), dried (sodium sulfate) and concentrated in vacuo. The resulting crude product, a white solid, was used in the next step without further purification (2.402 g, 94 %). HPLC (method LCMS_fastgradient) tR = 0.88 min. 1H NMR (CDCl3, 300 MHz): delta 4.52 (dt, J = 4.2, 12.6 Hz, 2 H), 6.16 (tt, J= 4.2, 54.8 Hz, 1 H). A solution (or suspension) of the appropriate aminoazole 1a-h (0.01 mol) in solvent (15 ml) (EtOH in the case of compound 3a, dioxane in the case of compounds 3b-h) was stirred at 50C and treated by adding 3,3-diethoxypropionitrile (4) (1.5 ml, 0.01 mol), then 36% HCl solution (0.86 ml, 0.01 mol). The reaction mixture was refluxed for 2.5-3 h, the suspension (or solution) was cooled to room temperature, and the target product was isolated by the method indicated for each particular compound.

In a 25 mL round-bottomed flask under N2 at 0 °C, sodium hydride (60percent in mineral oil, 0.059 g, 1.48 mmol) was added to a solution of 3,5-dimethyl-lH-l,2,4-triazole (0.112 g, 1.15 mmol) in DMF (5 mL) and the mixture was stirred for 30 min. The resulting mixture was then poured into H20 and the product was extracted with EtOAc (x3). The combined organic extract was washed with 0 (x3) and brine, dried over anhydrous sodium sulfate and concentrated to dryness. The residue was purified on ISCO using a 40 g column (0-5percent MeOH-DCM) to afford the title compound (0.248 mg, 0.675 mmol, 59percent yield) as a colorless gum. LC (Method B): 1.795 min. MS (APCI): calcd for Ci8H16BrN4 [M + H]+ m/z 367.1, found 367.0. H NMR (400 MHz, CDC13) delta ppm 7.66 (dd, J = 0.78, 1.57 Hz, 1H), 7.61 – 7.63 (m, 2H), 7.51 – 7.56 (m, 2H), 7.28 – 7.31 (m, 2H), 5.29 (s, 2H), 2.42 (s, 3H), 2.38 (s, 3H).
