N-doped carbon nanopipe-arrayed mesoporous carbon (N-CMK-5) with double-pore system, large surface area (992 m(2)/g), and high nitrogen amount (5.2 at.%) was synthesized by a simplified template route. In this route, mesoporous silica was directly used instead of using time-consuming aluminosilicate as a template and the carbonization process occurred in a nitrogen atmosphere instead of a vacuum. Furthermore, CMK-3, N-doped CMK-3, and CMK-5 ordered mesopomus carbons (OMCs) were compared with the as-prepared N-doped CMK-5 (N-CMK-5) to investigate the effect of microstructure on the electrochemical activity of OMCs. N-CMK-5 exhibited higher electrochemical activity than the other OMCs because it has more exposed defective sites and nitrogen active species. The developed N-CMK-5-based electrochemical sensor showed good practical application for the electrochemical detection of pesticide amitrole in real water sample. This work contributes a simplified route to synthesize tubular N-CMK-5 carbon and provides valuable information on the design of high-performance carbon electrode materials.

The construction materials that contacted with unsymmetrical dimethylhydrazine and the desorption solutions obtained when treating the contaminated surface of metals and alloys with water and reagents were studied by chromatography and mass spectrometry. Neutralization of unsymmetrical dimethylhydrazine was studied using ozone and shungite. Ozonation makes it possible to destroy the toxicant molecules chemically and physically adsorbed on the surface of metal constructions, due to which they can be reused and utilized. Shungite effectively adsorbs and catalytically decomposes not only unsymmetrical dimethylhydrazine and its transformation products, but also oligomer compounds formed during the storage of hydrazine fuel. Ozonation of spent shungite can increase the efficiency of destructive processes and completeness of its regeneration.

Hydrogen peroxide is a damage signal at sites of chronic inflammation. The question arises whether platelet-rich fibrin (PRF), platelet-poor plasma (PPP), and the buffy coat can neutralize hydrogen peroxide toxicity and thereby counteract local oxidative stress. In the present study, gingival fibroblasts cells were exposed to hydrogen peroxide with and without lysates obtained from PRF membranes, PPP, heated PPP (75 degrees C for 10 min), and the buffy coat. Cell viability was examined by trypan blue staining, live-dead staining, and formazan crystal formation. Cell apoptosis was assessed by cleaved caspase-3 Western blot analysis. Reverse transcription-quantitative polymerase chain reaction (RT-PCR) was utilized to determine the impact of PRF lysates on the expression of catalase in fibroblasts. It was reported that lysates from PRF, PPP, and the buffy coat-but not heated PPP-abolished the hydrogen peroxide-induced toxicity in gingival fibroblasts. Necrosis was confirmed by a loss of membrane integrity and apoptosis was ruled out by the lack of cleavage of caspase-3. Aminotriazole, an inhibitor of catalase, reduced the cytoprotective activity of PRF lysates yet blocking of glutathione peroxidase by mercaptosuccinate did not show the same effect. PRF lysates had no impact on the expression of catalase in gingival fibroblasts. These findings suggest that PRF, PPP, and the buffy coat can neutralize hydrogen peroxide through the release of heat-sensitive catalase.

1,3-Disulfonic acid imidazolium trifluoroacetate ([Dsim][CF3CO2]) was employed as an efficient, homogeneous and recyclable ionic liquid catalyst for the synthesis of triazoloquinazolinones, chromeno[4,3-d]benzothiazolopyrimidines and benzoimidazopyrimidine derivatives via one-pot multi-component reactions under thermal and solvent-free conditions. The catalyst is demonstrated excellent catalytic properties with good yields in short reaction times and low cost. All products were obtained by recrystallization and there was no need to tedious work-up.

The utility of the enaminonitriles 3a and 3b for the synthesis of the pyrazole derivatives 5a,b, 7a,b, diaminopyrimidine derivatives 9,11, pyrazolo[1,5-alpha]pyrimidines 12,15, triazolo[4,3-alpha]pyrimidines 13,16, imidazo[1,2-alpha]pyrimidine derivatives 14 and 17, was explored. Most of the synthesized compounds showed excellent in vitro antitumor activity against MCF-7 cell line. They also exhibted high antimicrobial and antioxidant activities. Density functional theory (DFT) calculations at the B3LYP/6-31G level of theory have been carried out to investigate the equilibrium geometry of the novel phenylpyrazolo[1,5-alpha]pyrimidin-6-yl)methanone derivative 12 and phenylpyrazolo[1,5-alpha]pyrimidine derivative 15. The structure-activity relationship (SAR) has been used to correlate the biological activity with the appropriate quantum such as total energy. Hepatocyte cell death is a key process in the pathogenesis of nonalcoholic steatohepatitis (NASH). However, the factors responsible for and mechanisms underlying NASH-related cell death have not yet been elucidated in detail. We herein investigated the effects of intracellular glyceraldehyde (GA)-derived advanced glycation end-products (AGEs), named toxic AGEs (TAGE), on the production of reactive oxygen species (ROS), which have been implicated in the pathogenesis of NASH. Cell death related to intracellular TAGE accumulation was eliminated in the hepatocyte carcinoma cell line HepG2 by the antioxidant effects of N-acetyl-L-cysteine. The intracellular accumulation of TAGE increased ROS production and the expression of Nrf2, including its downstream gene. These results suggest that ROS are produced in association with the accumulation of TAGE and are a direct trigger for cell death. We also investigated the factors responsible for these increases in ROS. Catalase activity did not decrease with the accumulation of TAGE, while mitochondrial membrane depolarization was enhanced in cells treated with GA. These results indicate that TAGE play an important role in mitochondrial abnormalities and increases in ROS production, both of which are characteristic features of NASH. The suppression of TAGE accumulation has potential as a new therapeutic target in the progression of NASH.

In the present study, a rapid, less expensive, clean and environmental friendly route to synthesis new pyrazoles, pyrazolopyridazines and condensed pyrimidines was developed via grinding of 2-(3-(dimethylamino)acryloyl)-3H-benzo[f]chromen-3-one (1) with different reagents. All the new compounds were characterized and established using elemental analysis and spectral data. Eight compounds were selected for in vitro antiproliferative against different human cancer cell lines entitled melanoma, cancers of the lung, leukemia, breast, brain, colon, prostate, ovary and kidney by the USA NCI. Sirtuin 3 (SIRT3) is a deacetylase important for antioxidant protection, cell longevity, and aging. We hypothesized that SIRT3 improve oxidative resistance of aged cells and improve cell therapy in aged patients. In vitro, the proliferation and oxidative resistance of human mesenchymal stem cells (hMSCs) significantly declined with age. The expression and activity of antioxidant enzymes, including catalase (CAT) and manganese superoxide dismutase (MnSOD), increased after transfection of SIRT3 in hMSCs from older donors (O-hMSCs). The protein level of Forkhead box O3a (FOXO3a) in nucleus increased after SIRT3 overexpression. The antioxidant capacity of O-hMSCs increased after SIRT3 overexpression. 3-Amino-1,2,4-triazole (3-AT, CAT inhibitor) or diethyldithiocarbamate (DETC, SOD inhibitor) that was used to inhibit CAT or SOD activity significantly blocked the antioxidant function of SIRT3. When two inhibitors were used together, the antioxidant function of SIRT3 almost disappeared. Following myocardial infarction and intramyocardial injections of O-hMSCs in rats in vivo, the survival rate of O-hMSCs increased by SIRT3 transfection. The cardiac function of rats was improved after SIRT3-overexpressed O-hMSC transplantation. The infarct size, collagen content, and expression levels of matrix metalloproteinase 2 (MMP2) and MMP9 decreased. Besides, the protein level of vascular endothelial growth factor A and vascular density increased after cell transplantation with SIRT3-modified O-hMSCs. These results indicate that damage resistance of hMSCs decline with age and SIRT3 might protect O-hMSCs against oxidative damage by activating CAT and MnSOD through transferring FOXO3a into nucleus. Meanwhile, the therapeutic effect of aged hMSC transplantation can be improved by SIRT3 overexpression.

A series of novel 2-(5-aminomethylene-4-oxo-2-thioxothiazolidin-3-yl)-3-phenylpropionic acid ethyl esters has been synthesized. Target compounds were evaluated for their trypanocidal activity towards Trypanosoma brucei brucei and Trypanosoma brucei gambiense. Several hit-compounds (8, 10, 12) inhibited growth of the parasites at sub-micromolar concentrations (IC50 0.027-1.936 mu M) and showed significant selectivity indices (SI= 108-1396.2) being non-toxic towards the human primary fibroblasts. The screening of anticancer activity in vitro within NCI DTP protocol allowed to identify active 2-(5-{[5-(2,4-dichlorobenzyl)-thiazol-2-ylamino]methylene}- 4-oxo-2-thioxothiazolidin-3-yl)-3-phenylpropionic acid ethyl ester 14 that demonstrated inhibition against all 59 human tumor cell lines with the average GI(50) value of 2.57 mu M.
