Amitrole is reported as a novel kind of transparent poly(vinyl chloride) (PVC) thermal stabilizer that has both excellent inhibition of zinc burning thermal stability and antibacterial ability in this paper, which provide the possibility of applying the whole PVC industry into medical and food packaging, artificial leather, wire and cable, indoor decoration, and so on. From the results of Congo red test, discoloration test, and dynamic thermal stability test, we could clearly find out that amitrole could effectively replace the unstable chlorine atom and inhibit zinc burning of transparent PVC, while the long-term stability should be improved. Next, the stabilization mechanism of amitrole was also confirmed. The antibacterial activity of amitrole was measured by the bacteriostatic ring experiments. It was observed that the PVC films had good antibacterial properties after modified by amitrole. At last, optimal design of the amitrole/ZnSt(2) (1:1)/ESBO formulation was studied, which have strong potential as thermal stabilizer to prepare super transparency and antibacterial PVC resin.

Mechanisms controlling the expression of the electrically conductive pili (e-pili) of Geobacter species are of interest because of the important role of e-pili in diverse biogeochemical processes, anaerobic digestion and electromicrobiological applications. We investigated the function of the protein, designated Spc (short pilin chaperone), encoded by the gene immediately downstream from the gene for PilA, the monomer that assembles into e-pili. Multiple lines of evidence suggest that Spc forms an oligomer that is associated with the inner membrane. Mutating the start codon of spc to prevent translation increased the transcript abundance of pilA but greatly diminished the abundance of PilA, and e-pili could no longer be detected. Cross-linking, protein capture and two-hybrid studies demonstrated that Spc and PilA interacted. Two sites in PilA for electrostatic interaction with Spc were identified. The results demonstrate that Spc is required for PilA stability prior to incorporation into e-pili, suggesting that Spc has a chaperone function that may be specific to the relatively short PilA monomers that assemble into e-pili. These results are important for identifying microorganisms likely to express e-pili from (meta)genomic data and for the construction of microbial strains expressing e-pili.

Conjugated polymers are promising light harvesters for water reduction/oxidation due to their simple synthesis and adjustable bandgap. Herein, both cyanamide and triazole functional groups are first incorporated into a heptazine-based carbon nitride (CN) polymer, resulting in a mesoporous conjugated cyanamide-triazole-heptazine polymer (CTHP) with different compositions by increasing the quantity of cyanamide/triazole units in the CN backbone. Varying the compositions of CTHP modulates its electronic structures, mesoporous morphologies, and redox energies, resulting in a significantly improved photocatalytic performance for both H(2)and O(2)evolution under visible light irradiation. A remarkable H(2)evolution rate of 12723 mu mol h(-1)g(-1)is observed, resulting in a high apparent quantum yield of 11.97% at 400 nm. In parallel, the optimized photocatalyst also exhibits an O(2)evolution rate of 221 mu mol h(-1) g(-1), 9.6 times higher than the CN counterpart, with the value being the highest among the reported CN-based bifunctional photocatalysts. This work provides an efficient molecular engineering approach for the rational design of functional polymeric photocatalysts.

The cullin-RING ubiquitin ligases (CRLs) are responsible for about 20% of cellular protein degradation and regulate diverse cellular processes, and the dysfunction of CRLs is implicated in human diseases. Targeting the CRLs has become an emerging strategy for the treatment of human diseases. Herein, we describe the discovery of a hit compound from our in-house library and further structure-based optimizations, which have enabled the identification of new triazolo[1,5-a]pyrimidine-based inhibitors targeting the DCN1-UBC12 interaction. Compound WS-383 blocks the DCN1-UBC12 interaction (IC50 = 11 nM) reversibly and shows selectivity over selected kinases. WS-383 exhibits cellular target engagement to DCN1 in MGC-803 cells. WS-383 inhibits Cul3/1 neddylation selectively over other cullins and also induces accumulation of p21, p27, and NRF2. Collectively, targeting the DCN1-UBC12 interaction would be a viable strategy for selective neddylation inhibition of Cul3/1 and may be of therapeutic potential for disease treatment in which Cul3/1 is dysregulated.

The layer-by-layer assembly of molecules is ubiquitous in nature. Highly ordered structures formed in this manner often exhibit fascinating material properties. A layer hydrogen bonding pairing approach allows the development of tunable energetic materials with targeted properties. A series of unusual energetic compounds based on 1H,1 ‘ H-5,5 ‘-bistetrazole-1,1 ‘-diolate (1), such as the salts of 3-amino-1,2,4-triazolium (2), aminoguanidinium (3), and hydrazinium (4), and the cocrystals of 4-amino-1H-pyrazole (5), 2-methylimidazole (6), and imidazole (7), were synthesized using this strategy. The structures of the obtained products 2-7 were fully characterized by elemental analysis, IR spectroscopy, H-1 NMR and C-13 NMR spectroscopy, and single-crystal X-ray analysis. Their thermal decomposition behavior was studied by differential scanning calorimetry and thermogravimetry. Their mechanical sensitivities and detonation performances were also analyzed in detail. Results show that products 2-7 exhibit higher density, better detonation performances, and more excellent sensitivities than those of the same species of cation salts previously reported.

Grapevine (Vitis vinifera L.) is a crop of major economic importance. However, grapevine yield is guaranteed by the massive use of pesticides to counteract pathogen infections. Under temperate-humid climate conditions, downy mildew is a primary threat for viticulture. Downy mildew is caused by the biotrophic oomycete Plasmopara viticola Berl. & de Toni, which can attack grapevine green tissues. In lack of treatments and with favourable weather conditions, downy mildew can devastate up to 75% of grape cultivation in one season and weaken newly born shoots, causing serious economic losses. Nevertheless, the repeated and massive use of some fungicides can lead to environmental pollution, negative impact on non-targeted organisms, development of resistance, residual toxicity and can foster human health concerns. In this manuscript, we provide an innovative approach to obtain specific pathogen protection for plants. By using the yeast two-hybrid approach and the P. viticola cellulose synthase 2 (PvCesA2), as target enzyme, we screened a combinatorial 8 amino acid peptide library with the aim to identify interacting peptides, potentially able to inhibit PvCesa2. Here, we demonstrate that the NoPv1 peptide aptamer prevents P. viticola germ tube formation and grapevine leaf infection without affecting the growth of non-target organisms and without being toxic for human cells. Furthermore, NoPv1 is also able to counteract Phytophthora infestans growth, the causal agent of late blight in potato and tomato, possibly as a consequence of the high amino acid sequence similarity between P. viticola and P. infestans cellulose synthase enzymes.

This study investigated the nutrient content and reuse potential of wastewater generated during hydrothermal liquefaction of microalgal biomass. The hydrothermal liquefaction reaction was tested at 270, 300, 330, and 345 degrees C to determine the effect of temperature on the formation of non-biodegradable dissolved organic nitrogen (nbDON). Total nitrogen, ammonium, color, and toxicity were selected as key characteristics for the reuse of hydrothermal liquefaction wastewater. Results indicated that a higher concentration of nbDON(5) (nbDON defined with a 5 day growth assay) and more diverse heterocyclic N-containing organic compounds were associated with greater toxicity as measured by a growth rate assay. For the tested temperature ranges, the total nitrogen content of the hydrothermal liquefaction wastewater slightly decreased from 5020 +/- 690 mg L-1 to 4160 +/- 120 mg L-1, but the % nbDON(5) fraction increased from 57 +/- 3 %DON to 96 +/- 5 %DON. The temperature of hydrothermal liquefaction reactions can be optimized to maximize carbon conversion and nitrogen recovery. (C) 2019 Elsevier Ltd. All rights reserved.

The Mbp1 protein functions as a DNA-binding protein in the MluI cell cycle box-binding complex and plays significant roles in yeast development. In this study, an ortholog of yeast Mbp1, BbMbp1, was characterized in a filamentous insect mycopathogen, Beauveria bassiana. BbMbp1 plays an important role in morphological changes under aerial and liquid environments. On the aerial surface, BbMbp1 was indispensable for the biogenesis of conidiophores and conidiation. Under submerged conditions, the increment BbMbp1 mutant displayed abnormal spore-producing structures, with a dramatic decrease in blastospore yield (similar to 95%). The virulence of the increment BbMbp1 mutant was notably weakened, which might be due to the defect in in vivo blastospore formation in the insect. Moreover, disruption of BbMbp1 resulted in a substantial reduction in hyphal growth on cadavers. Comparative transcriptomics revealed that BbMbp1 mediated different transcriptomes during the formation processes of conidia and blastospores. Yeast one-hybrid assays demonstrated that BbMbp1 was required for transcriptional control of a cell wall protein gene, BbCwp, and an integral membrane protein gene, BbImp that played significant roles in conidiation and blastospore formation respectively. Porous coordination polymers (PCPs) of Zn2+, oxalic acid and mixture ratio of 3-amino, 1,2,4-triazole (Ataz) and 1,2,4-triazole (Taz) were synthesized to capture of carbon dioxide. A series of molar fraction Taz/(Taz + ATaz) prepared as 0.1;0.3;0.4;0.5;0.6;0.7 and 0.9 and also only ATaz and Taz ligand as 0 and 1 molar fraction respectively. Mixture of Taz and ATaz crystal induce new structure of 0.4, 0.5 and 0.6 molar fraction. Nitrogen adsorption of 50% mixture each ligand contribute the highest surface area as function of optimum condition to integrate pore space and amine group presence with adsorbed as 290 mgg(-1). This phenomena also shown through CO2 adsorption amount as 135 mgg(-1), instead of 0.4, 0.6 and 0.7 molar fraction observed higher amount of CO2 compared with only Taz ligand (X=1), indicate the integrated effects both of the ligand could generated the capture of higher amount of carbon dioxide.

Water is essential for honey bees (Apis mellifera L.), but contaminated sources of water in agricultural environments represent a risk of exposure to potentially harmful contaminants. Providing clean water to honey bees could be an efficient and cost-effective measure for beekeepers to reduce bee mortality associated with pesticides and improve the health of their colonies. The main goal of this study was to design a waterer prototype to fulfill the water requirements of honey bees and to evaluate the potential of this waterer in improving colonies’ health in agricultural settings, through mitigating the possible impact of an exposure to pesticides from puddle water. We tested the preference of honey bees regarding water composition and waterer prototypes, among which honey bees showed a strong preference for salted water and a poultry-type waterer. Our waterer models were quickly adopted and intensively used through the season in both the context of honey production in field crops and pollination services in cranberry crops. However, in neither context did the use of waterers reduce worker mortality nor increase overall colony weight. Our waterers provided bees with water containing fewer pesticides and were associated with reduced risks of drowning compared to natural sources of water. Our study suggests that the use of waterers fulfills an important requirement for honey bees and represents an interesting and convenient precautionary measure for beekeepers.
