Novel bis[(thiazol-2-yl)acetonitrile] derivatives were prepared in good yields by the cyclocondensation of bis(bromoacetyl) derivatives with two equivalents of 2-cyanothioacetamide in dioxane at reflux. The bis[(thiazol-2-yl)acetonitrile] derivatives were taken as synthetic precursors for the synthesis of novel bis(aminoazolo[1,5-a]pyrimidines), bearing thiazole moiety. The target molecules were prepared by the three component one pot reaction of bis[(thiazol-2-yl)acetonitrile] derivatives, dimethylformamide-dimethylacetal and several of 3-aminopyrazoles in pyridine under microwave irradiation at 140 degrees C for 2 h. Using the same protocol, novel bis(aminotriazolo[1,5-a]pyrimidines), bis(aminopyrimido[1,2-a]benzimidazoles) and bis(aminopyrido [1,2-a]benzimidazoles), incorporating thiazole moieties, were prepared by using the appropriate heterocyclic amine or 2-(1H-benzoimidazol-2-yl)acetonitrile instead of 3-aminopyrazoles. The structure of the newly prepared thiazoles was confirmed via considering their elemental analyses and spectral data.

Insulin resistance is thought to be a common link between obesity and Non-Alcoholic Fatty Liver Disease (NAFLD). NAFLD has now reached epidemic status worldwide and identification of molecules or pathways as newer therapeutic strategies either to prevent or overcome insulin resistance seems critical. Dysregulated hepatic lipogenesis (DNL) is a hallmark of NAFLD in humans and rodents. Therefore, reducing DNL accretion may be critical in the development of therapeutics of NAFLD. In our in vivo model (high-fat-diet fed [HFD] obese mice) we found Zinc oxide nanoparticles (ZnO NPs) significantly decreased HFD-induced hepatic steatosis and peripheral insulin resistance. This protective mechanism of ZnO NPs was signaled through hepatic SIRT1-LKB1-AMPK which restricted SREBP-lc within the cytosol limiting its transcriptional ability and thereby ameliorating HFD mediated DNL. These observations indicate that ZnO NP can serve as a therapeutic strategy to improve the physiological homeostasis during obesity and its associated metabolic abnormalities. (C) 2019 Elsevier Inc. Heteroatom-doped ordered mesoporous carbons (OMCs) have currently been considered as promising electrode materials for electrochemical sensors due to the combined advantages of ordered mesoporous materials and heteroatom-doped carbon materials. Herein, a novel nitrogen and sulfur co-doped OMCs (N,S-OMC) has been prepared via a nanocasting strategy with an inexpensive methylene blue as single precursor. The obtained mesoporous carbon has platelet morphology, short mesoporous channel together with a large surface area (549 m(2)/g) as well as rich N- and S-containing functional groups (6.8 at.% N and 2.3 at.% S). Compared with the graphene (GR) and carbon nanotube (CNT) electrode material, the N,S-OMC exhibited a higher electrochemical activity towards the oxidation of herbicide amitrole, ascribable to N,S-OMC’s open mesoporous structures and abundant electroactive defect sites on the carbon skeleton. And, an amitrole electrochemical sensor with N,S-OMC modified electrode as working electrode was fabricated, exhibiting a good selectivity, stability, reproducibility and wide linear range of 3-750 mu M. Moreover, the N,S-OMC-based electrochemical sensor was proved feasible in river water sample analyses, showing a satisfied recovery ranging from 97.03% to 105.42%. The results not only demonstrate cheap methylene blue can be used as single precursor for the N,S-OMC preparation, but also confirm the N,S-OMC is promising in amitrole sensor fabrication.

The widely expressed G-protein coupled receptors (GPCRs) are versatile signal transducer proteins that are attractive drug targets but structurally challenging to study. GPCRs undergo a number of conformational rearrangements when transitioning from the inactive to the active state but have so far been believed to adopt a fairly conserved inactive conformation. Using (FNMR)-F-19 spectroscopy and advanced molecular dynamics simulations we describe a novel inactive state of the adenosine 2A receptor which is stabilised by the aminotriazole antagonist Cmpd-1. We demonstrate that the ligand stabilises a unique conformation of helix V and present data on the putative binding mode of the compound involving contacts to the transmembrane bundle as well as the extracellular loop 2.

Background: Advanced glycation end products (AGEs) are known to be involved in the pathophysiology of diabetic complications, neurodegenerative diseases, and aging. Preventing the formation of AGEs can be helpful in the management of these diseases. Objectives: Two classes of previously synthesized traizole Schiff’s bases (4H-1,2,4-triamle-4-Schiff’s bases 1-14, and 4H-1,2,4-triazole-3-Schiff’s bases 15-23) were evaluated for their in vitro antiglycation activity. Methods: In vitro fructose-mediated human serum albumin (HSA) glycation assay was employed to assess the antiglycation activity of triazole Schiff’s bases. The active compounds were subjected to cytotoxicity analysis by MTT assay on mouse fibroblast (3T3) cell line. Molecular docking and simulation studies were carried out to evaluate the interactions and stability of compounds with HSA. Anti-hyperglycemic and antioxidant activities of selected non-cytotoxic compounds were evaluated by in vitro a-glucosidase inhibition, and DPPH free radical scavenging assays, respectively. Results: Compound 1 (IC50 =47.30 +/- 0.38 mu M) from 4H-1,2,4-triazole-4-Schiff’s bases has exhibited antiglycation activity comparable to standard rutin (IC50 =54.5 +/- 0.05 mu M) along with a stable RMSD profile in MD simulation studies. Compound 1 also exhibited a potent a-glucosidase inhibitory activity, and moderate antioxidant property. Other derivatives showed a weak antiglycation activity with IC50 values between 248.1-637.7 mu M. Compounds with potential antiglycation profile were found to be non-cytotoxic in a cellular assay. Conclusion: The study identifies triazole Schiff s bases active against fructose-mediated glycation of HSA, thus indicates their potential against late diabetic complications due to production of advancedend products (AGEs).

A thermo-responsive copolymer with a gas-switchable LCST-type phase transition was synthesized by reversible addition-fragmentation chain transfer (RAFT) polymerization of N-isopropylacrylamide (NIPAM) and glycidyl methacrylate (GMA), followed by post-polymerization functionalization with sodium 3-amino-1,2,4-triazole (ATANa). Incorporating ionic moieties provides an elevated cloud point of 61 degrees C. Importantly, both CO2 and SO2 cause a reduction in the cloud point of the polymer solution. The CO2-triggered system can be easily and fully recovered to its initial state by introducing an inert gas (e.g. N-2), whereas the SO2-triggered system shows only partial recovery. The pH-dependent phase transition behaviors confirm that the gas bubbling-induced pH changes contribute to the gas-switchable cloud point of P(NIPAM-co-(GMA-ATANa)). In addition, P(NIPAM-co-(GMA-ATA)) (ATA: 3-amino-1,2,4-triazole), a counterpart of P(NIPAM-co-(GMA-ATANa)), was prepared and the relevant solution phase transition behavior was studied. Its cloud point shift in response to gas bubbling and pH adjustment is opposite to that of P(NIPAM-co-(GMA-ATANa)), indicating that the latter does not result from the protonation of amidine groups. Alternatively, a reversible H+-induced decrease in hydrophilicity was thus proposed. This contribution enriches the family of thermo-responsive polymers by introducing gas-sensitive polyelectrolytes and also broadens the scope of gas-responsive smart materials.

In this study, 2-urea-chitosan oligosaccharide derivatives (2-urea-COS derivatives) and 2,6-diurea-chitosan oligosaccharide derivatives (2,6-diurea-COS derivatives) were successfully designed and synthesized via intermediate 2-methoxyformylated chitosan oligosaccharide. All samples were characterized and compared based on FT-IR, H-1 NMR spectroscopy, and elemental analysis. The antifungal effects of COS derivatives were tested against Fusarium oxysporum f. sp. niveum, Phomopsis asparagus, and Botrytis cinereal. Their antioxidant properties, including superoxide radicals’ scavenging activity, hydroxyl radicals’ scavenging activity, and DPPH radicals’ scavenging activity were also explored within different concentrations. COS derivatives bearing urea groups showed improved bioactivity compared with pristine COS and 2,6-diurea-COS derivatives had a higher biological activity than 2-urea-COS derivatives in tested concentrations. Additionally, L929 cells were used to carry out cytotoxicity test of COS and COS derivatives by CCK-8 assay. The results indicated that some of samples showed low cytotoxicity. These findings offered a suggestion that COS derivatives bearing urea groups are promising biological materials.

Ultrathin C3N5 nanosheets with enhanced photocatalytic methylene blue (MB) degradation and H-2-evolution performance were prepared from thermal treatment of 3-amino-1,2,4-triazole (3-AT) and NH4Cl followed with a protonate procedure. The characterization results revealed that the protonating process could contribute to the exfoliation of C3N5 with large surface area, the effective charge transfer capability and the modified band structure. The as-prepared C3N5 nanosheets exhibited enhanced properties in photocatalytic reactions such as MB photodegradation and H-2-evolution from water splitting. This study offered a feasible route to prepare highly-efficient two-dimensional photocatalyst, which could be applied potentially for implementation in wide range of energy generation and environmental applications.

In this work, the coordination-based energy sacrificial bonds have been constructed in the interphase between lignin and polyolefin elastomer to prepare high performance lignin-based thermoplastic elastomers (TPEs). The strength and toughness of lignin-based TPEs can be adjusted by choosing different nitrogen heterocyclic compounds as reactive assistants and Fe3+ or Zr2+ as metal coordination centers. It was demonstrated that 3-Amino-1,2,4-triazole with three nitrogen atoms in the heterocydic ring and one nitrogen branch chain could form the most efficient coordination bond system and generate the best mechanical performance. The system with ferric iron as coordination center exhibited better enhancement effect than divalent zinc. By adjusting the nitrogen-containing reactive additives or metal salts as coordination centers, the mechanical performance of the lignin-based TPE can be regulated, which provides a method for making green bio-composites with good strength and toughness, and also promotes the high value utilization of lignin in polymer materials. Under the catalysis of solid acidic ion exchange resin Amberlyst-15, using ethanol as solvent, aromatic aldehyde, beta-ketoester, and 3-amino-1,2,4-triazole or 2-aminobenzo as starting materials, a series of triazolo[1,5-a]pyrimidine derivatives 5a similar to 5m and dihydrobenzo[4,5]imidazo[1,2-a]pyrimidines 6a similar to 6g were synthesized by three-component one-pot reaction of imidazole derivatives. The method is simple and efficient, and has short reaction time, simple post-treatment and wide application range of the substrate. The catalyst can be recycled for 4 times without the significant decrease of catalytic activity. It conforms to the basic standard of green chemistry.
