Heat stress (HS) has serious effects on plant development, resulting in heavy agricultural losses. A critical transcription factor network is involved in plant adaptation to high temperature. DEHYDRATION RESPONSIVE ELEMENT-BINDING PROTEIN2A (DREB2A) is a key transcription factor that functions in plant thermotolerance. The DREB2A protein is unstable under normal temperature and is degraded by the 26S proteasome; however, the mechanism by which DREB2A protein stability dramatically increases in response to HS remains poorly understood. In this study, we found that the DREB2A protein of Arabidopsis (Arabidopsis thaliana) is stabilized under high temperature by the posttranslational modification SUMOylation. Biochemical data indicated that DREB2A is SUMOylated at K163, a conserved residue adjacent to the negative regulatory domain during HS. SUMOylation of DREB2A suppresses its interaction with BPM2, a ubiquitin ligase component, consequently increasing DREB2A protein stability under high temperature. In addition, analysis of plant heat tolerance and marker gene expression indicated that DREB2A SUMOylation is essential for its function in the HS response. Collectively, our data reveal a role for SUMOylation in the maintenance of DREB2A stability under high temperature, thus improving our understanding of the regulatory mechanisms underlying HS response in plant cells. SUMOylation increases the protein stability of DREB2A, a key transcription factor in the response to heat stress in plant cells.

The mechanisms of binary catalyst therapy (BCT) and photodynamic therapy (PDT) are based on the formation of reactive oxygen species (ROS). This ROS formation results from specific chemical reactions. In BCT, light exposure does not necessarily initiate ROS formation and BCT application is not limited to regions of tissues that are accessible to illumination like photodynamic therapy (PDT). The principle of BCT is electron transition, resulting in the interaction of a transition metal complex (catalyst) and substrate molecule. Mn-III-tetraphenylporphyrin chloride (MnClTPP) in combination with an ascorbic acid (AA) has been proposed as an appropriate candidate for cancer treatment regarding the active agents in BCT. The goal of this study was to determine whether MnClTPP in combination with AA would be a promising agent for BCT. The problem of used MnClTPP’s, low solubility in water, was solved by MnClTPP loading into PLGA matrix. H2O2 produced during AA decomposition oxidized MnClTPP to high-reactive oxo-Mn-V species. MnClTPP in presence AA leads to the production of excessive ROS levels in vitro. ROS are mainly substrates of catalase and superoxide dismutase (H2O2 and O-2(center dot-)). SOD1 and catalase were identified as the key players of the MnClTPP ROS-induced cell defense system. The cytotoxicity of MnClTPP-loaded nanoparticles (NPs) was greatly increased in the presence of specific catalase inhibitor (3-amino-1,2,4-triazole (3AT)) and superoxide dismutase 1 (SOD1) inhibitor (diethyldithio-carbamate (DDC)). Cell death resulted from the combined activation of caspase-dependent (caspase 3/9 system) and independent pathways, namely the AIF translocation to nuclei. Preliminary acute toxicity and in vivo anticancer studies have been revealed the safe and potent anticancer effect of PLGA-entrapped MnClTPP in combination with AA. The findings indicate that MnClTPP-loaded PLGA NPs are promising agents for BCT.

Ultrathin C3N5 nanosheets with enhanced photocatalytic methylene blue (MB) degradation and H-2-evolution performance were prepared from thermal treatment of 3-amino-1,2,4-triazole (3-AT) and NH4Cl followed with a protonate procedure. The characterization results revealed that the protonating process could contribute to the exfoliation of C3N5 with large surface area, the effective charge transfer capability and the modified band structure. The as-prepared C3N5 nanosheets exhibited enhanced properties in photocatalytic reactions such as MB photodegradation and H-2-evolution from water splitting. This study offered a feasible route to prepare highly-efficient two-dimensional photocatalyst, which could be applied potentially for implementation in wide range of energy generation and environmental applications.

Bismuth oxyhalides (BiOX (X = Cl, Br, I) are considered to be an important p-type semiconductors in the photocatalysis applications. In particular, tetragonal BiOBr is considered as a stable photocatalyst due to its resilient absorption in the visible region with an band gap energy of 2.8 eV. In the meantime, lanthanide ions (with 3+ oxidation state) implies as conversion catalyst gained huge impact and remain a serious topic in materials chemistry. Here we synthesized upconversion photocatalyst mainly consists of BiOBr with the Er 3+ and Tb 3+ ions along with low band gap g-C3N5 for the improved photocatalytic performances. The synthesized Er3+/ Tb3+@BiOBr-g-C3N5 heterojunction was systematically characterized by XRD, and FT-IR for the confirmation of the composite and their morphology were analysed with FESEM and HR-TEM analysis which revealed that the sheets of g-C3N5 were decorated by Er3+/Tb3+ loaded BiOBr microspheres. The XPS analysis confirmed the suitable oxidation state of all the individual elements existing in the composite. As the UV-DRS analysis showed that the band gap of the Er3+/Tb3+ BiOBr-gC3N5 heterojunction was narrowed to 2.64 eV. To evaluate the photocatalytic efficiency of the synthesized g-C3N5, Er3+/Tb3+@BiOBr and Er3+/Tb3+@BiOBr-gC3N5 heterojunction under the simulated visible light irradiation source towards the aqueous sulfamethoxazole degradation. The Er3+/Tb3+@BiOBr-gC3N5 heterojunction shows maximum degradation efficiency of 94.2% after 60 min of visible light irradiation whereas the pure g-C3N5 provided about 43.8% and Er3+/Tb3+@BiOBr implies 55.2% degradation efficiency. The plausible degradation mechanism of pollutant removal was proposed.

The rice receptor-like cytoplasmic kinase 185 (OsRLCK185) interacts with the chitin receptor complex OsCERK1/CEBiP and positively regulates chitin-induced immune responses including MAP kinase activation, ROS production and defense gene expression. To elucidate the regulatory mechanisms of OsRLCK185-mediated immunity, we searched for interactors of OsRLCK185. OsDRE2a, rice homologs of the yeast Dre2 protein, were identified as novel interactors of OsRLCK185. OsDRE2a interacted with OsRLCK185 at plasma membrane. The conserved cysteine residues in CIAPIN1 domain of OsDRE2a were essential for tight interaction of OsRLCK185. OsDRE2a was phosphorylated by OsRLCK185. The expression of OsDRE2a and OsDRE2b was induced after chitin treatment. Reduction of OsDRE2a and OsDRE2b mRNA levels by RNA interference resulted in the decreased chitin-induced ROS production. Thus, it is likely that OsDRE2 regulates OsRLCK185-mediated immune responses.

Cobalt oxide (Co3O4) nanoparticles decorated on mesoporous carbon nitride (Co3O4/MCN) nanocomposites for photocatalytic hydrogen evolution were investigated in this work. MCN was prepared using 3-amino-1,2,4-triazole, high nitrogen content, as a single molecular carbon and nitrogen precursor and SiO2 nanoparticles as the hard template. Complementary characterization techniques were employed to understand the textural and chemical properties of the nanocomposites. The bare MCN showed high photocatalytic activity under visible light irradiation without using any co-catalyst. The photocatalytic activity of Co3O4/MCN with a Co3O4 mass content of 5 wt % presented two times higher than the bare MCN, which is attributed to the enhanced visible-light harvesting and more efficient charge separation. Mechanistic study shows lower electron-hole recombination rate, higher charge separation efficiency occurs after the formation of p-n type heterojunction. (C) 2019 Hydrogen Energy Publications LLC. Grapevine (Vitis vinifera L.) is a crop of major economic importance. However, grapevine yield is guaranteed by the massive use of pesticides to counteract pathogen infections. Under temperate-humid climate conditions, downy mildew is a primary threat for viticulture. Downy mildew is caused by the biotrophic oomycete Plasmopara viticola Berl. & de Toni, which can attack grapevine green tissues. In lack of treatments and with favourable weather conditions, downy mildew can devastate up to 75% of grape cultivation in one season and weaken newly born shoots, causing serious economic losses. Nevertheless, the repeated and massive use of some fungicides can lead to environmental pollution, negative impact on non-targeted organisms, development of resistance, residual toxicity and can foster human health concerns. In this manuscript, we provide an innovative approach to obtain specific pathogen protection for plants. By using the yeast two-hybrid approach and the P. viticola cellulose synthase 2 (PvCesA2), as target enzyme, we screened a combinatorial 8 amino acid peptide library with the aim to identify interacting peptides, potentially able to inhibit PvCesa2. Here, we demonstrate that the NoPv1 peptide aptamer prevents P. viticola germ tube formation and grapevine leaf infection without affecting the growth of non-target organisms and without being toxic for human cells. Furthermore, NoPv1 is also able to counteract Phytophthora infestans growth, the causal agent of late blight in potato and tomato, possibly as a consequence of the high amino acid sequence similarity between P. viticola and P. infestans cellulose synthase enzymes.

Porous liquids (PLs), an emerging kind of liquid materials with permanent porosity, have attracted increasing attention in gas capture. However, directly turning metal-organic frameworks (MOFs) into PLs via a covalent linkage surface engineering strategy has not been reported. Additionally, challenges including reducing the cost and simplifying the preparation process are daunting. Herein, we proposed a general method to transform Universitetet i Oslo (UiO)-66-OH MOFs into PLs by surface engineering with organosilane (OS) and oligomer species via covalent bonding linkage. The oligomer species endow UiO-66OH with superior fluidity at room temperature. Meanwhile, the resulting PLs showed great potential in both CO2 adsorption and CO2/N-2 selective separation. The residual porosity of PLs was verified by diverse characterizations and molecular simulations. Besides, CO2 selective capture sites were determined by grand canonical Monte Carlo (GCMC) simulation. Furthermore, the universality of the covalent linkage surface engineering strategy was confirmed using different classes of oligomer species and another MOF (ZIF-8-bearing amino groups). Notably, this strategy can be extended to construct other PLs by taking advantages of the rich library of oligomer species, thus making PLs promising candidates for further applications in energy and environment-related fields, such as gas capture, separation, and catalysis.

A series of novel inulin derivatives were designed and synthesized by the introduction of amino heterocyclic moieties onto carboxymethyl inulin with the aid of 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride and N-hyd roxysucci ni mide. The target products were prepared via three – step chemical synthesis, and structures were identified by FTIR and H-1 NMR spectroscopy. Antioxidant activities of inulin derivatives including DPPH – radical scavenging assay, superoxide – radical scavenging assay, hydroxyl – radical scavenging assay, and reducing power were estimated. Meanwhile, their antifungal activities, including Colletotrichum lagenarium and Bottytis cinerea, were also explored by hyphal measurement. In particular, inulin derivatives bearing heterocyclic moieties exhibited a remarkable improvement over inulin on antioxidant and antifungal activities, and their bioactivities decreased roughly in the order of 2ATCMI > 4APCMI > 3APCMI > 2APCMI > 3ATCMI > CMI > inulin. Furthermore, the cytotoxicities of inulin derivatives against L929 cells were evaluated by CCK-8 in vitro, and all samples showed weak cytotoxicities. In a nutshell, the paper provides a practical approach to synthesize novel inulin derivatives with dramatically enhanced bioactivity and good biocompatibility. N-doped carbon nanopipe-arrayed mesoporous carbon (N-CMK-5) with double-pore system, large surface area (992 m(2)/g), and high nitrogen amount (5.2 at.%) was synthesized by a simplified template route. In this route, mesoporous silica was directly used instead of using time-consuming aluminosilicate as a template and the carbonization process occurred in a nitrogen atmosphere instead of a vacuum. Furthermore, CMK-3, N-doped CMK-3, and CMK-5 ordered mesopomus carbons (OMCs) were compared with the as-prepared N-doped CMK-5 (N-CMK-5) to investigate the effect of microstructure on the electrochemical activity of OMCs. N-CMK-5 exhibited higher electrochemical activity than the other OMCs because it has more exposed defective sites and nitrogen active species. The developed N-CMK-5-based electrochemical sensor showed good practical application for the electrochemical detection of pesticide amitrole in real water sample. This work contributes a simplified route to synthesize tubular N-CMK-5 carbon and provides valuable information on the design of high-performance carbon electrode materials.
