It is desirable yet challenging to synthesize a highly ordered holey carbon nitride material with excess nitrogen atoms for a sustainable solar H(2)evolution. Herein, we report the development of directed highly ordered nitrogen-rich honeycomb-like mesoporous carbon nitride nanosheets by using a novel synthetic approach for this purpose. The unique formation route, surface structure and charge carrier dynamics of the two dimensional holey nanosheets are comprehensively monitored and confirmed by SAXS, HRTEM, AFM, BET, XPS and TRPL analyses. Non-stoichiometric high nitrogen content mesoporous nanosheets with the final stoichiometry of g-C(3)N(4.5)acquiring a high specific surface area (382 m(2)g(-1)), remarkable pore size (7.2 nm) and sheet thickness similar to 5-6 nm are realized in this first report. This elegant material possesses unique low band-gap energy (2.42 eV). Remarkably, the as-synthesized g-C(3)N(4.5)NSs exhibit a record high photocatalytic H(2)evolution rate of 8180 mu mol g(-1)h(-1)under the present light irradiation (420 <=lambda <= 510 nm) condition. The apparent quantum efficiency is found to be as high as 27.14% at 420 nm and retains its photocatalytic activity for longer consecutive catalytic cycles. Bigger pore size and pore volume, and thin walls leading to shortening of the path length of exciton pairs, efficient charge separation and the prolonged average life time of the charge carriers, and lone electron pairs associated with the excess nitrogen content are found to be the reasons behind the excellent visible light driven water splitting reaction. The widely expressed G-protein coupled receptors (GPCRs) are versatile signal transducer proteins that are attractive drug targets but structurally challenging to study. GPCRs undergo a number of conformational rearrangements when transitioning from the inactive to the active state but have so far been believed to adopt a fairly conserved inactive conformation. Using (FNMR)-F-19 spectroscopy and advanced molecular dynamics simulations we describe a novel inactive state of the adenosine 2A receptor which is stabilised by the aminotriazole antagonist Cmpd-1. We demonstrate that the ligand stabilises a unique conformation of helix V and present data on the putative binding mode of the compound involving contacts to the transmembrane bundle as well as the extracellular loop 2.

The isoindolinone and biaryl scaffolds are prevalent in natural products and drug molecules, which have showed broad and interesting biological activities. The efficient construction of such hybridized molecules and biological evaluation are of great interest to medicinal chemistry community. In this communication, we report an efficient Bronsted acid-promoted C(sp(3))-H functionalization approach that enables the rapid construction of biologically important isoindolinone/[1,2,4]triazolo[1,5-a]pyrimidine hybrids from 5-methyl-7-(2,4,6-trimethoxyphenyl)-[1,2,4]triazolo[1,5-a]pyrimidine, 2-formylbenzoic acid and various anilines. The title compounds were generated in high to excellent yields (up to 96%) regardless of the electronic nature and steric effects of the substituents. In this reaction, an isoindolinone scaffold, one C-C single bond, and two C-N bonds were formed simultaneously with high atom economy. In this work, we have envisioned that the methyl group linked to the electron-deficient N-heterocycles could be used as a new synthetic handle for late-state diversification and may have broad applications in the field of organic and medicinal chemistry. Besides, the title compounds have exhibited promising activity against the SKP2-CKS1 interaction. It is still a great challenge to prepare lignin/polyolefin composites with good strength and toughness. Inspired by the energy sacrificial mechanism from biomaterials, dynamic hydrogen bonds (H-bonds) are incorporated into the lignin/ethylene-propylene-diene monomer (EPDM) composite system to improve the interfacial interactions between lignin and EPDM. Such H-bonding interactions are demonstrated to be able to simultaneously improve the modulus, strength, and toughness of lignin/EPDM composites without sacrificing the extensibility. This work provides a facile method for the high-valued utilization of lignin in the preparation of high-performance elastomer composites using bio-renewable resources as reinforcing agent.

1-(1,2,4-Triazol-3-yl)-1H-tetrazole (trt) and 1-(1-tert-butyl-1,2,4-triazol-3-yl)-1H-tetrazole (btrt) were synthesized by heterocyclization of corresponding amines with triethyl orthoformate and sodium azide. The interaction of trt with copper(II) chloride in ethanol in the presence of hydrochloric acid was found to result in 10 coordination polymer [CuCl2(trt)(H2O)](n)center dot nH(2)O. N-tert-butylated ligand btrt reacts with the copper salt in ethanol giving 2D coordination polymer [CuCl2(btrt)](n). Tetrazole ring-opening reaction with the formation of [CuCl2(3-amino-1,2,4-triazolium)(2)][CuCl4]center dot 2H(2)O was observed in the interaction of trt with copper(II) chloride in aqueous hydrochloric acid. The temperature-dependent magnetic susceptibility measurements of [CuCl2(btrt)](n) revealed that the copper(II) ions were weakly ferromagnetically coupled showing a coupling constant J of 1.2 cm(-1). Based on the extensive biological activities of thiazole derivatives against different types of diseases, we are interested in the effective part of many natural compounds, so we synthesized a new series of compounds containing di-, tri- and tetrathiazole moieties. The formation of such derivatives proceeded via reaction of 2-bromo-1-(4-methyl-2-(methylamino)thiazol-5-yl)ethan-1-one with heterocyclic amines, o-aminothiophenol and thiosemicarbazone derivatives. The structure and mechanistic pathways for all products were discussed and proved based on spectral results, in addition to conformational studies. Our aim after the synthesis is to investigate their antimicrobial activity against various types of bacteria and fungi species. Preceeding such an investigation, a molecular docking study was carried out with selected conformers, as representative examples, against three pathogen-proteins. This preliminary stage could support the biological application. The potency of these compounds as antimicrobial agents has been evaluated. The results showed that derivatives which have di- and trithiazole rings displayed high activity that exceeds the used standard antibiotic.

In search of unprecedented tri and/or tetrapod pharmacophoric conjugates, a series of 32 new 4-ethyl-1H-benzo[b][1,4]diazepin-2(3H)-ones were synthesized and properly elucidated using MS, IR, NMR, and elemental analysis. In vitro investigation of 11 compounds of this series, using a panel of two human tumor cell lines namely; human breast adenocarcinoma (MCF-7), and human colorectal carcinoma (HCT-116), revealed promising cytotoxic activities. Among all synthesized compounds, analogue 9 displayed maximum cytotoxicity with IC50 values of 16.19 +/- 1.35 and 17.16 +/- 1.54 mu M against HCT-116 and MCF-7, respectively, compared to standard drug doxorubicin.

Graphene nanomaterials have attracted a great interest during the last years for different applications, but their possible impact on different biological systems remains unclear. Here, an assessment to understand the toxicity of commercial polycarboxylate functionalized graphene nanoplatelets (GN) on the unicellular fungal model Saccharomyces cerevisiae was performed. While cell proliferation was not negatively affected even in the presence of 800 mg L-1 of the nanomaterial for 24 hours, oxidative stress was induced at a lower concentration (160 mg L-1), after short exposure periods (2 and 4 hours). No DNA damage was observed under a comet assay analysis under the studied conditions. In addition, to pinpoint the molecular mechanisms behind the early oxidative damage induced by GN and to identify possible toxicity pathways, the transcriptome of S. cerevisiae exposed to 160 and 800 mg L-1 of GN was studied. Both GN concentrations induced expression changes in a common group of genes (337), many of them related to the fungal response to reduce the nanoparticles toxicity and to maintain cell homeostasis. Also, a high number of genes were only differentially expressed in the GN800 condition (3254), indicating that high GN concentrations can induce severe changes in the physiological state of the yeast.

The efficient removal of radioactive iodine (I-2) has drawn worldwide attention because of the severe effects of radiotoxicity on the environment and human health. As a result, efficient porous materials for the capture and safe storage of radioactive iodine are required. In this work, nanoscale zeolitic imidazolate framework-8 (ZIF-8) and its amino-functionalized ZIF-8 derivatives with 3-amino-1,2,4-triazole (Atz), denoted as ZIF8-A, were prepared via postsynthetic modification, and their performance for I-2 removal from n-hexane was investigated. The abundant amine functionalities in the ZIF8-A structure including the primary and secondary amine groups (-NH2 and -NH) and triazole-ring (-C=N-NH-) not only serve as effective adsorption sites but also form strong interactions with I-2 via electron transfer from the nitrogen atom of the ligand to I-2. The adsorption isotherms and the kinetics were well fitted by the Langmuir model and the pseudo-second-order kinetic model, respectively, suggesting a monolayer chemisorption process. ZIF8-A61 achieved an adsorption capacity of more than 870% compared to the pristine ZIF-8. Based on the experimental and computational results, a plausible mechanism for the adsorption of I-2 on ZIF8-A nanocrystals is proposed by considering the host-guest interactions, specifically, those between the amine functional groups and I-2. In addition, the reusability and structural stability of the adsorbents were investigated.

Breast cancer risk from pesticides may be missed if effects on mammary gland are not assessed in toxicology studies required for registration. Using US EPA’s registration documents, we identified pesticides that cause mammary tumors or alter development, and evaluated how those findings were considered in risk assessment. Of 28 pesticides that produced mammary tumors, EPA’s risk assessment acknowledges those tumors for nine and dismisses the remaining cases. For five pesticides that alter mammary gland development, the implications for lactation and cancer risk are not assessed. Many of the mammary-active pesticides activate pathways related to endocrine disruption: altering steroid synthesis in H295R cells, activating nuclear receptors, or affecting xenobiotic metabolizing enzymes. Clearer guidelines based on breast cancer biology would strengthen assessment of mammary gland effects, including sensitive histology and hormone measures. Potential cancer risks from several common pesticides should be re-evaluated, including: malathion, triclopyr, atrazine, propylene oxide, and 3-iodo-2-propynyl butylcarbamate (IPBC).
