Large amounts of effectors are secreted by the oesophageal glands of plant-parasitic nematodes, but their molecular mode of action remains largely unknown. We characterized aMeloidogyne incognitaprotein disulphide isomerase (PDI)-like effector protein (MiPDI1) that facilitates nematode parasitism. In situhybridization showed thatMiPDI1was expressed specifically in the subventral glands ofM. incognita. It was significantly upregulated during parasitic stages. Immunolocalization demonstrated MiPDI1 secretionin plantaduring nematode migration and within the feeding cells. Host-induced silencing of theMiPDI1gene affected the ability of the nematode to infect the host, whereasMiPDI1expression inArabidopsisincreased susceptibility toM. incognita, providing evidence for a key role of MiPDI1 inM. incognitaparasitism. Yeast two-hybrid, bimolecular fluorescence complementation and coimmunoprecipitation assays showed that MiPDI1 interacted with a tomato stress-associated protein (SlSAP12) orthologous to the redox-regulated AtSAP12, which plays an important role in plant responses to abiotic and biotic stresses.SAP12silencing or knocking out inNicotiana benthamianaand Arabidopsis increased susceptibility toM. incognita. Our results suggest that MiPDI1 acts as a pathogenicity factor promoting disease by fine-tuning SAP-mediated responses at the interface of redox signalling, defence and stress acclimation in Solanaceae andArabidopsis.

Porous coordination polymers (PCPs) of Zn2+, oxalic acid and mixture ratio of 3-amino, 1,2,4-triazole (Ataz) and 1,2,4-triazole (Taz) were synthesized to capture of carbon dioxide. A series of molar fraction Taz/(Taz + ATaz) prepared as 0.1;0.3;0.4;0.5;0.6;0.7 and 0.9 and also only ATaz and Taz ligand as 0 and 1 molar fraction respectively. Mixture of Taz and ATaz crystal induce new structure of 0.4, 0.5 and 0.6 molar fraction. Nitrogen adsorption of 50% mixture each ligand contribute the highest surface area as function of optimum condition to integrate pore space and amine group presence with adsorbed as 290 mgg(-1). This phenomena also shown through CO2 adsorption amount as 135 mgg(-1), instead of 0.4, 0.6 and 0.7 molar fraction observed higher amount of CO2 compared with only Taz ligand (X=1), indicate the integrated effects both of the ligand could generated the capture of higher amount of carbon dioxide.

Graphene nanomaterials have attracted a great interest during the last years for different applications, but their possible impact on different biological systems remains unclear. Here, an assessment to understand the toxicity of commercial polycarboxylate functionalized graphene nanoplatelets (GN) on the unicellular fungal model Saccharomyces cerevisiae was performed. While cell proliferation was not negatively affected even in the presence of 800 mg L-1 of the nanomaterial for 24 hours, oxidative stress was induced at a lower concentration (160 mg L-1), after short exposure periods (2 and 4 hours). No DNA damage was observed under a comet assay analysis under the studied conditions. In addition, to pinpoint the molecular mechanisms behind the early oxidative damage induced by GN and to identify possible toxicity pathways, the transcriptome of S. cerevisiae exposed to 160 and 800 mg L-1 of GN was studied. Both GN concentrations induced expression changes in a common group of genes (337), many of them related to the fungal response to reduce the nanoparticles toxicity and to maintain cell homeostasis. Also, a high number of genes were only differentially expressed in the GN800 condition (3254), indicating that high GN concentrations can induce severe changes in the physiological state of the yeast.

Phenacylbromide derivatives constitute a multilateral group of precursors for the synthesis of numerous heterocycles of organic compounds. Briefly, 5-(2-bromo-acetyl)-substituted-thiophene derivative has been used as a synthon for synthesis of new thiophene-containing compounds through the reaction with nucleophilic nitrogen compounds and thioamides. The suggested structures of the newly synthesized thiophene compounds were confirmed and assured with different spectroscopic tools and with CHN elemental analysis. Additionally, the antimicrobial activity of these thiophene compounds was recorded to investigate their potency against various types of bacteria and fungi. Results showed that these compounds exhibit significant inhibitory activity against the growth of tested bacterial and fungal strains and that some derivatives were more potent than the employed reference drugs.

A thermo-responsive copolymer with a gas-switchable LCST-type phase transition was synthesized by reversible addition-fragmentation chain transfer (RAFT) polymerization of N-isopropylacrylamide (NIPAM) and glycidyl methacrylate (GMA), followed by post-polymerization functionalization with sodium 3-amino-1,2,4-triazole (ATANa). Incorporating ionic moieties provides an elevated cloud point of 61 degrees C. Importantly, both CO2 and SO2 cause a reduction in the cloud point of the polymer solution. The CO2-triggered system can be easily and fully recovered to its initial state by introducing an inert gas (e.g. N-2), whereas the SO2-triggered system shows only partial recovery. The pH-dependent phase transition behaviors confirm that the gas bubbling-induced pH changes contribute to the gas-switchable cloud point of P(NIPAM-co-(GMA-ATANa)). In addition, P(NIPAM-co-(GMA-ATA)) (ATA: 3-amino-1,2,4-triazole), a counterpart of P(NIPAM-co-(GMA-ATANa)), was prepared and the relevant solution phase transition behavior was studied. Its cloud point shift in response to gas bubbling and pH adjustment is opposite to that of P(NIPAM-co-(GMA-ATANa)), indicating that the latter does not result from the protonation of amidine groups. Alternatively, a reversible H+-induced decrease in hydrophilicity was thus proposed. This contribution enriches the family of thermo-responsive polymers by introducing gas-sensitive polyelectrolytes and also broadens the scope of gas-responsive smart materials.

Recently, activated carbon adsorption for water treatment regained substantial attention due to the emerging task to remove trace organic compounds such as pesticides. In many applications, especially in decentralized water treatment, one major drawback of adsorbents is their limited recyclability due to inadequate logistics or uneconomical reactivation. In this lab-scale study, we present the temperature swing adsorption in the aqueous phase that allows the in situ regeneration of fixed-bed adsorbers, and prove its technical feasibility. Complying with circular water economy principles, we eliminated the pivotal need for regular replacement and consumables by employing only clean water instead of dedicated regeneration solutions. Adsorption of the herbicide amitrole in aqueous solution on granular activated carbon was exothermic (Delta H = -14.4 +/- 3.2 kJ mol(-1) for T = 20-94 degrees C) and followed the Freundlich model. The proposed method consisting of a short counterflow flush with liquid water at 125 degrees C effectively regenerated the adsorbent. Hence, we obtained a cyclic steady state operation with breakthrough after 122 +/- 14 bed volumes (at c(out)/c(in) = 0.2), cycle-average rejection of 90 +/- 1%, and water recovery of up to 78 +/- 4%. No thermal aging of adsorbent was observed over the investigated 17 cycles.

Two-hybrid systems can be used for investigating protein-protein interactions and may provide important information about gene products with unknown function. Despite their success in mapping protein interactions, two-hybrid systems have remained mostly untouched by improvements in next-generation DNA sequencing. The two-hybrid systems rely on one-versus-all methods in which each bait is sequentially screened against an entire library. Here, we developed a screening method that joins both bait and prey as a convergent fusion into one bacterial plasmid vector that can then be amplified and paired-end sequencing by next-generation sequencing (NGS). Our method enables all-versus-all sequencing (AVA-Seq) and utilizes NGS to remove multiple bottlenecks of the two-hybrid system. AVA-Seq allows for high-resolution protein-protein interaction mapping of a small set of proteins and has the potential for lower-resolution mapping of entire proteomes. Features of the system include ORF selection to improve efficiency, high bacterial transformation efficiency, a convergent fusion vector to allow paired-end sequencing of interactors, and the use of protein fragments rather than full-length proteins to better resolve specific protein contact points. We demonstrate the system’s strengths and limitations on a set of proteins known to interact in humans and provide a framework for future large-scale projects.

8-Acetyl-7-hydroxy-4-methyl coumarin and 3-amion-1,2,4-triazole were used to prepare a Schiff base (Sbat). The [Ag(Sbat)(NO3)]center dot H2O and [Cu(Sbat)(OH)(H2O)(2)]center dot 3H(2)O complexes were separated by the reaction of the Schiff base with Ag(i) and Cu(ii) metal ions. All the prepared compounds were subjected to elemental and spectral analyses (IR, UV-vis, H-1 NMR, and mass spectroscopy). Moreover, the complexes were also subjected to TG analysis, molar conductance measurements, magnetic moment analysis, and XRD measurements. The value of the molar conductance designates their non-electrolytic character. The local minimum structure, global properties and nonlinear optical parameters of the Schiff base were calculated at the B3LYP/6-31++(d,p) level of theory and those of its complexes were calculated at the B3LYP/GENECP level. The electronic spectra in the UV-visible region of the Schiff base (Sbat) and [Ag(Sbat)(NO3)]center dot H2O and [Cu(Sbat)(OH)(H2O)(2)]center dot 3H(2)O complexes in DMSO as a solvent were investigated experimentally and theoretically using the time dependent density functional theory (TD-DFT) method at the CAM-B3LYP/6-311++G(d,p) level of theory. The studied compounds were screened for their in vitro antibacterial activity against three Gram-positive (B. subtilis, S. aureus and S. faecalis) and three Gram-negative bacteria (E. coli, N. gonorrhoeae and P. auregenosa). An antifungal assay was performed against two fungal (C. albicans and A. flavus) species. The Ag-Sbat complex showed the highest antimicrobial activity. The cytotoxic activity of the Schiff base and its silver complex was tested in vitro against the MCF-7 (breast cancer) and HCT-116 (colon cancer) cell lines.

Amitrole is reported as a novel kind of transparent poly(vinyl chloride) (PVC) thermal stabilizer that has both excellent inhibition of zinc burning thermal stability and antibacterial ability in this paper, which provide the possibility of applying the whole PVC industry into medical and food packaging, artificial leather, wire and cable, indoor decoration, and so on. From the results of Congo red test, discoloration test, and dynamic thermal stability test, we could clearly find out that amitrole could effectively replace the unstable chlorine atom and inhibit zinc burning of transparent PVC, while the long-term stability should be improved. Next, the stabilization mechanism of amitrole was also confirmed. The antibacterial activity of amitrole was measured by the bacteriostatic ring experiments. It was observed that the PVC films had good antibacterial properties after modified by amitrole. At last, optimal design of the amitrole/ZnSt(2) (1:1)/ESBO formulation was studied, which have strong potential as thermal stabilizer to prepare super transparency and antibacterial PVC resin.

Fast and accurate practical sample detection is a great challenge in on-site detection. Herein, we developed a CoFe2O4@HNTs/ AuNPs substrate for rapid and efficient magnetic solid-phase extraction (MSPE) surface-enhanced Raman scattering (SERS) detection of aromatic amines and nitrofuran in real samples all-in-one. Magnetic CoFe2O4 beads filled inside halloysite nanotubes (HNTs) can avoid aggregation of particles, endow the substrate with the rapid magnetic separation ability to simplify the pretreatment procedure, and reduce complex matrix interference. Meanwhile, outer surface AuNPs can generate electromagnetic enhancement and hot spots to amplify Raman signals of target molecules enriched/concentrated by HNTs. The CoFe2O4@HNTs/AuNPs substrate exhibited excellent SERS activity (high sensitivity, good reproducibility, and repeatability), pH stability (3.0-11.0), and good MSPE ability (fast magnetic enrichment/separation ability within 5 min). The CoFe2O4@HNTs/AuNPs MSPE SERS substrate can be applied for the determination of 4,4′-thioaniline and nitrofurantoin with a linear range of 0.054-21.7 mg/L and 0.05-1.0 mg/L, and the limits of detection were down to 0.026 mg/L and 0.014 mg/L, respectively. Furthermore, the enhancement factor (EF) of the substrate to 4,4′-thioaniline is up to 2.7 x 10(7). Besides, the substrate can realize practical SERS determination of trace 4,4-thioaniline in cosmetics and nitrofurantoin in fish feed and aquatic samples. The recoveries were varied from 71.6% to 103.6% for 4,4-thioaniline in hair dyes and 81.9% to 116.3% for nitrofurantoin in fish feed and aquatic samples, respectively. Such a robust and efficient MSPE SERS substrate possesses great potential in rapid detection (within 15 min) for a practical sample, and it also provides a methodology for the preparation of other HNTs-based composites.
