Gestational exposure to tebuconazole affects the development of rat fetal Leydig cells

Tebuconazole is a triazole fungicide, used in agriculture to treat phytopathogenic fungi, and as a biocide, has been reported to be related to reproductive and developmental toxicity. The purpose of this study was to investigate the effect of tebuconazole exposure on rat fetal Leydig cells and fetal testis during pregnancy. Isavuconazole Therapeutic Drug Monitoring during Long-Term Treatment for Chronic Pulmonary Aspergillosis

Isavuconazole is the newest triazole antifungal, and it displays a favorable pharmacokinetic and safety profile. Less is known about its long-term use in immunocompetent hosts. We performed a retrospective service evaluation of isavuconazole therapeutic drug monitoring in patients with chronic pulmonary aspergillosis. Adverse events (AEs) and dose adjustments made during routine clinical practice were recorded, and AEs were classified based on Common Terminology Criteria for Adverse Events v5.0. Forty-five patients (mean age, 64 years) had 285 isavuconazole blood drug levels measured (mean level, 4.1 mg/liter). A total of 117 measurements (41%) were performed on patients on a 100-mg daily dose instead of 200 mg, and all had blood levels of >1 mg/liter. Age (P = 0.012) and a daily dose of 200 mg versus 100 mg (P = 0.02) were independent predictors of levels of >6 mg/liter. AEs were recorded for 25 patients (56%). The mean drug level at the first measurement was 5.5 +/- 2 mg/liter for patients reporting AEs, compared with 4.2 +/- 1.7 mg/liter for those not reporting AEs (P = 0.032). The cutoff threshold best predictive of an AE was 4.6 mg/liter (area under the concentration-time curve, 0.710). Sixteen patients (36%) discontinued isavuconazole therapy due to AEs. Twenty-six patients (58%) continued on isavuconazole beyond 6 months. Asthma (P = 0.022) and a daily dose of 200 mg versus 100 mg (P = 0.048) were associated with AEs of grade 2 or higher. A reduced daily dose (100 mg versus 200 mg) of isavuconazole resulted in satisfactory drug levels in a substantial number of patients; it was better tolerated and enabled continuation of therapy for prolonged periods.

Co-crystals are emerging as members of supramolecular family having particular practical and fundamental significance for chemists, crystallographers, pharmaceutical scientists, and theoreticians. The presented study is focused to synthesize anti-cancer co-crystals of commercially available nandrolone (1), a synthetic anabolic-androgenic steroidal drug. Co-crystallization is done using economical and green grinding and reflux methods to obtain nandrolone (Nan): salicylic acid (Sal) and nandrolone (Nan): 3-amino-1,2,4-triazole (Triz) co-crystals, in 2:1 and 1:1 stoichiometric ratios, respectively. The structural analysis and characterization were carried out using single-crystal X-ray diffraction, and vibrational spectroscopy. Nandrolone (1) crystallizes in monoclinic P2(1) space group, while the co-crystal-I (Nan:Sal) and co-crystal-II (Nan:Triz), co-crystallized in the orthorhombic P2(1)2(1)2(1) space group. The intermolecular hydrogen bonds O-H center dot center dot center dot O, C-H center dot center dot center dot O, N-H center dot center dot center dot O, O-H center dot center dot center dot N, and N-H center dot center dot center dot N between the active pharmaceutical ingredient (nandrolone) and co-formers (salicylic acid, and 3-amino-1,2,4-triazole) stabilize the structures of cocrystals. Synthesis and energetic properties of homocubane based high energy density materials

The energetic properties of homocubane derivatives have been evaluated for the first time. The newly synthesized compounds have been fully characterized by spectroscopic methods and single crystal X-ray analysis. Detailed computational studies carried out using the B3LYP/6-311++G(d,p) level of theory reveal that these compounds have higher densities and higher heat of formation as compared to common hydrocarbon fuels. The calculated performances of these compounds in terms of their ballistic properties, particularly density specific impulse (rho I-sp), in liquid and in solid propellant systems are substantially superior to those of the conventional fuel RP1 and binder HTPB, respectively. TGA-DTG analysis confirmed that most of these compounds are thermally stable with high onset temperatures. Most of the compounds, except oxadiazole, triazole and triazine containing cages, are also kinetically stable in terms of their HOMO-LUMO energy gap. Quite remarkably, three compounds emerged as excellent candidates for volume-limited applications owing to their high density, density specific impulse as well as kinetic and thermodynamic stabilities.

Herein, this work presents a novel chiral tetrakis-iodo-triazole structure as a neutral halogen bond donor for both chiral anion-recognition and enantioinduction in ion-pair organocatalysis. NMR-titration studies revealed significant differences in anion affinity between the halogen bonding receptor and its hydrogen bonding parent. Selective recognition of chiral dicarboxylates and asymmetric induction in a benchmark organocatalytic reaction were demonstrated using the halogen bond donor. Inversions in the absolute sense of chiral recognition, enantioselectivity, and chiroptical properties relative to the related hydrogen donor were observed. Computational modeling suggested that these effects were the result of distinct anion-binding modes for the halogen- versus hydrogen-bond donors.

Design, Synthesis, Evaluation and Molecular Docking Studies of Novel Triazole Linked 1,4-Dihydropyridine-isatin Scaffolds as Potent Anticancer Agents

A series of novel triazole linked isatin-dihydropyridine hybrids (N1-N15) have been synthesized and examined for their anti proliferative activity against human cancer cell lines viz. HeLa, Huh-7, PC-3, IMR-32 and MCF-7. All of the synthesized hybrids have shown moderate to potent cytotoxicity against all the tested cell lines except IMR-32. Compounds N1, N2 and N13 have displayed an enhanced inhibitory potency against Huh-7 cell line as compared to the standard drug, doxorubicin. Out of the three, N2 has shown the highest in vitro inhibitory action with IC50 values of 6.73 +/- 0.33 mu M and 17.94 +/- 0.23 mu M against Huh-7 and MCF-7 cell lines, respectively. The docking studies of these most potent compounds have also been investigated which identified that N2 might be an excellent drug-like candidate worthy of further pursuit.
