The DCM phase was dried over anhydrous Na2S04, concentrated under reduced pressure and chromatographed on silica gel using MeOH and DCM (8:92) as eluents to get the desired amide 8 (305.3 mg, 90 % yield) as a white solid compound. -NMR (400 MHz, DMSO-de): <514.80-14.01 (br s, 1H), 9.25 (br s, 1H), 8.50 (br s, 1H), 7.44 (d, J= 8.4 Hz, 2H), 7.32 (d, J= 8 Hz, 2H), 4.47 (d, J= 6 Hz, 2H); 19F -NMR (376 MHz, DMSO-de): <5 - 56.89 ppm. The DCM phase was dried over anhydrous Na2S04, filtered and concentrated under reduced pressure and chromatographed on silica gel using EtOAc as eluents to get the desired amide 10 (62.8 mg, 24 % yield) as a white solid compound. -NMR (400 MHz, DMSO-de): <514.66 (br s, 1H), 9.23 (br s, 1H), 8.49 (br s, 1H), 7.60-7.58 (m, 2H), 7.22 (d, J= 8.4 Hz, 1H), 4.43 (d, J= 6 Hz, 2H), 3.86 (s, 3H); 19F -NMR (376 MHz, DMSO-de): <5 -60.82 ppm. To a reaction of 1/7-1, 2, 4-triazole-3 -carboxylic acid (226 mg, 2.0 mmol) and 3,4- dimethoxyaniline (322 mg, 2.1 mmol) in dry DMF (10 mL) was added 1- hydroxybenzotriazole monohydrate (337 mg, 2.2 mmol) and EDC (N-(3- dimethylaminopropyl)-N’-ethylcarbodiimide) (422 mg, 2.2 mmol) followed by N- methylmorpholine (0.88 mL, 8.0 mmol via syringe). The mixture was stirred at room temperature under nitrogen and the solids were gradually dissolved. The contents were stirred at room temperature for 24.0 h, and then slowly diluted into ice cold water and extracted with DCM (50 mL x 2). The DCM phase was washed with cold water (100 mL x 2). The DCM phase was dried over anhydrous Na2S04, filtered and concentrated under reduced pressure and then chromatographed on silica gel using EtOAc as eluent to get the desired amide 1 (60 mg, 12% yield) as a light brown solid. -NMK (400 MHz, DMSO- de): <514.79 (br s, 1H), 10.34 (br s, 1H), 8.58 (br s, 1H), 7.55-7.44 (m, 2H), 6.94-6.93 (m, 1H), 3.76-3.74 (m, 6H) ppm. It was stirred further for 20min at room temperature at which time LC/MS analysis confirmed full consumption of starting material. This crude compound was submitted to the analytical group for purification to afford the desired product.
