After the resulting mixture was cooled to 25 to 35 C., water and toluene were added. After the phases were separated, the aqueous phase was washed with toluene 3 to 5 times. The combined organic phases were concentrated under a maximum temperature of 70 C. The ethyl group of the thioester (1 mol) was removed by treated the resulting residue with sodium methoxide (approximately 8 mol) in dimtheylformamide (300 mL). The reaction mixture was stirred under nitrogen over night at 120 C. After cooling to room temperature, the reaction was quenched by adding methyl iodide (5 mol) under constantly stirring. Then the mixture was poured into water (350 mL) and extracted with tert-butyl methyl ether (100 mL*3). The organic phase was washed with saturated sodium sulphate solution and condensed. The residue was dissolved in warm toluene and washed with dilute hydrochloric acid aqueous solution, up to two times. After filtration of the precipitate and crystallization from isopropanol/Isopropyl ether (8/2, v/v), the D form of Chemical Structure A where X is -SH was obtained in ~65% yield (Chemical Structure 9).

To a solution of 1,2,4-triazole, 1 (27.6 g, 399.6 mmol) in dichloromethane (80 mL) was simultaneously added bromine in dichloromethane (50%v/v, 80 mL) and an aqueous solution of sodium hydroxide (50% w/v,96 mL). The rate of addition was such that the temperature of the reaction mass was maintained between 10-15C. After the complete addition, the temperature was raised to 25C and the stirring was continued for an additional 12 h. Into this was then added 6 N aq.HCl and stirring continued for 1 h.The solid thus precipitated was filtered, and dried under vacuum at 50C to afford 2 as an off-white solid(80 g, 90%). The product was used without further purification.

To a solution (3 ML) of 1,2,4-triazole (0.259 g, 3.75 mmol) in N,N-dimethylformamide was added small portions of sodium hydride (60% dispersion in oil, 0.12 g, 3.0 mmol) at about 20 C., and the mixture was stirred for about 3 hr until hydrogen was not generated.To a solution of sodium salt of 1,2,4-triazole thus obtained was added dropwise a solution (5.5 ML) of the total amount of (2R,3R)-3-(2′,4′-difluorophenyl)-3,4-epoxy-2-methanesulfonyoxybutane obtained above in N,N-dimethylformamide at room temperature.The mixture was stirred at 75-80 C. for 1.5 hr.The reaction mixture was added dropwise to water (20 ML), and the mixture was extracted 3 times with ethyl acetate (20 ML).The extracted ethyl acetate layers were mixed, and the mixture was washed twice with saturated brine (10 ML) and dried over anhydrous magnesium sulfate.After filtration, the filtrate (ethyl acetate solution) was concentrated and the obtained concentrate was subjected to silica gel column chromatography (SiO2, 5 g) and eluted with n-heptane-ethyl acetate (10:1)ethyl acetate.The objective fraction was concentrated to give a pale-yellow oil (0.297 g).The obtained pale-yellow oil was crystallized from a mixed solvent of ethyl acetate (1 ML)-n-heptane (4 ML) to give the title compound (0.185 g, yield from (2R)-2′,4′-difluoro-2-hydroxypropiophenone: 44%).As a result of analysis by HPLC, the optical purity was 100% e.e.
