The suspension was stirred at -40 C for 20 minutes, then a homogeneous solution of (4-chloro-2-methoxy-3-(4- (trifluoromethyl)benzyl)quinolii^ (500 mg, 1.05 mmol, Intermediate 63: step b) in 2 ml, TFIF was introduced. A dark brownish solution immediately resulted. The reaction mixture was allowed to warm gradually to 0 C over 25 minutes, then was quenched with aqueous H4CI solution. The aqueous portion was extracted with EtOAc (3 x 35 mL) and the combined organics were washed with brine, dried over MgS04, filtered and concentrated to dryness. The residue was purified by FCC (20% EtOAc-hexanes increasing to 50% EtOAc) to provide the title compound as a white solid. H NMR (500 MHz,CDC ) delta 8.08 (d, J —— 2.1 Hz, 1 H), 7.85 (d, J —— 8.7 Hz, IH), 7.61 – 7.46 (m, 3H), 7.39 (d, J —— 8.1Hz, 2H), 6.94 (s, IH), 4.81 (s, IH), 4.32 (s, 2H), 4.09 (s, 3H), 3.98 (s, 3H), 1.95 (s, 3H), 1.82 (s, 3H); MS (ESI): mass calc. for Chemical Formula: C27H23CIF3N5Q3; Exact Mass: 557.1, m z found, 557.9 (M+H).

A solution of 1-methyl-1H-1,2,3-triazole (1.47 g, 17.7 mmol, PCT Int. Appl. 2008098104) in 20 mL THF was cooled to -40 C. in a dry-ice/acetonitrile bath. n-Butyllithium (1.6 M in hexane, 10.2 mL, 16.3 mmol) was added dropwise via syringe and the mixture was stirred at -40 C. for 30 minutes. A solution of 1-methyl-1H-imidazole-5-carbaldehyde (1.50 g, 13.6 mmol) in 10 mL THF was then added and the mixture was stirred for 5 minutes, then was transferred to an ice/water bath. After 1 hour, the mixture was quenched by addition of saturated aqueous NH4Cl, diluted with water, and extracted twice with EtOAc. The aqueous phase, which contained the title compound, was then concentrated. The residue was purified by flash column chromatography (silica gel, gradient 3-10% MeOH-DCM) to afford the title compound as a light yellow foam.

The combined organies were washed with brine, dried over Na2S04, filtered and concentrated to provide a brown oil. Chromatography on silica gel (3% MeOH-DCM increasing to 5% MeOH-DCM) to provide the title compound as a faint amber solid. MS m/e558.2 [M+H]+. Intermediate 13: bis(1-methyl-1H-1,2,3-triazol-5-yl)methanoneA solution of 1 -methyl- lH-1, 2, 3-triazole (0.954 g, 11.4 mmol, prepared according to PCX Int. AppL, 2008098104) in THF (22 mL) was stirred at-70 C under argon while -BuLi (2.56 M in hexanes, 4.29 mL, 11.0 mmol) was added dropwise over 5 minutes. After stirring for another 5 minutes, a solution of ethyl mefhoxy(methyl)carbamate (0.665 g, 4.99 mmol) in THF (3 mL) wras added dropwise over 5 minutes. After stirring at ~-70 C for an additional 5 minutes, the cold bath was removed and the light slurry was allowed to warm to room temperature with stirring for 1 hour 20 minutes. The reaction was then quenched at room temperature with 5 M aqueous H4CI (3 mL) and the aqueous layer was extracted with THF (1 x 6 mL). The combined organic layers were dried (Na2S04), filtered, and concentrated. A portion of the residue was crystallized from -’30 mL toluene to provide, after washing the filter cake with ether (1 > 3 mL) and heptane (1 x 3 mL), to yield the title compound as blunt needles.

General procedure: 1-Methylimidazole (0.49 g, 6.05 mmol) was added to a stirred solution of Key Intermediate-1 (2.3 g, 4.03 mmol) in acetonitrile (25 mL) at RT. The reaction mixture was heated to 90 C for 16 h and concentrated under reduced pressure. The crude material was triturated with diethyl ether (2 x 10 mL) to afford a brown solid (2.4g) which was used in the next steps without further purification. M/z 616.38 (M)+ TFA: H2O (9:1, 20 mL) was added to the brown solid (2.4 g) at RT. The reaction mixture was stirred for 3 h and concentrated under reduced pressure. The resulting crude product was triturated with diethyl ether (2 x 5 mL) and dried under high vacuum. The crude product was purified by preparative HPLC affording the title product as off-white solid (515 mg, 30% over 2 steps).

The reaction mixture became a dark brown color and was allowed to warm gradually to room temperature. The mixture was stirred for 60 minutes at, room, temperature then quenched with aqueous NH4CI solution. The aqueous portion was extracted with EtOAc, 3 x 50 mL. The combined organics were washed with brine, dried over MgS04, filtered and concentrated to provide a brown oil. Chromatography on silica gel (1% MeOH-DCM increasing to 5% MeOH-DCM) provided the title compound as a light brown solid. Racemic (4~chloro-2-methoxy-3-(4~(trifluorometliyl)benzyl)quinohyl)rnethanol was separated into its individual enantiomers using the following conditions: Chiralcel OD, 20 uM (Diacel) using ethanol with 242 nM detection to give the first eluting enantiomer as Intermediate 84b and the second eluting enantiomer as Intermediate 84c.

To this was added 1-methyl-1H-1,2,3-triazole (20.8 mg, 0.250 mmol). The vial was again flushed with nitrogen. To this was added NMP (0.5 mL).
