A novel pyridinium salt derivative, 2,3,4,5,6-pentaphenyl-1-(1H-1,2,4-triazol-3-yl)pyridinium perchlorate (1) has been designed and synthesized by using pyrylium perchlorate and 3-amino-1H-1,2,4-triazole as ingredients, which can be exploited for the binding and specific sensing of anion via hydrogen bonds. Compound 1 exhibits intense blue emission in solution with maximum emission at 469 nm with a red shift relative to that (426 nm) of solid state. The luminescence intensity of 1 could be selectively and sensitivity quenched with acetate ion and fluorine ion among various anions including F-, Cl-, Br-, I-, AcO- and NO3-. Furthermore, the possible detection mechanism was investigated. This work could contribute a credible potential sensing application of 1,2,4-triazolyl substituted pentaphenylpyridinium salts towards the specific anions in biological and environmental concerns. The reactive co-polymer approach is one of the most promising techniques for the synthesis of functional polyolefins. Following this concept, 1-hexene and p-methylstyrene are co-polymerized in the presence of a generic Brookhart-type catalyst. The microstructures of the co-polymers imply the tendency of p-methylstyrene co-monomers to place at the end of the structural branches formed by the chain walking reaction. The molar masses of the co-polymers decrease, not only at higher levels of co-monomer but surprisingly by decreasing reaction temperature. A mechanism consisting of a highly stable eta(3) metal-benzyl intermediate, which is quantitatively approved by density functional theory calculations, can delicately justify all the aforementioned observations. A series of the produced co-polymers is selectively functionalized by maleic anhydride at the benzylic position of p-methylstyrene, under very mild reaction conditions. Such a reactive intermediate opens the path for the introduction of different types of functionalities in polyolefins. Namely, the grafted co-polymers were further functionalized by a triazole ring, which provides a transient supramolecular network through intermolecular hydrogen bonding.

Synthesis of some new heterocyclic ring systems incorporated pyrimidine and pyridine moieties starting from 1-(furan-2-yl)-3-(thiophen-2-yl) chalcone was achieved. The structure of the new compounds was interpreted by spectral studies and ESI-MS analysis. Antimicrobial investigations of the designated compounds were performed towards some harmful pathogenic microbes. Antimicrobial tests proved that compound 11 unveiled a greater antimicrobial activity than other designed compounds. Docking of compound 11 into active site of DNA gyrase B chain displayed binding-energy of -13.05 kJ mol(-1) and distance at 3.18 A degrees. Furthermore, docking investigation was approved for the goal compounds into DNA gyrase B chain and exhibiting binding energy extended from -13.05 to -20.48 kJ mol(-1).

The Mbp1 protein functions as a DNA-binding protein in the MluI cell cycle box-binding complex and plays significant roles in yeast development. In this study, an ortholog of yeast Mbp1, BbMbp1, was characterized in a filamentous insect mycopathogen, Beauveria bassiana. BbMbp1 plays an important role in morphological changes under aerial and liquid environments. On the aerial surface, BbMbp1 was indispensable for the biogenesis of conidiophores and conidiation. Under submerged conditions, the increment BbMbp1 mutant displayed abnormal spore-producing structures, with a dramatic decrease in blastospore yield (similar to 95%). The virulence of the increment BbMbp1 mutant was notably weakened, which might be due to the defect in in vivo blastospore formation in the insect. Moreover, disruption of BbMbp1 resulted in a substantial reduction in hyphal growth on cadavers. Comparative transcriptomics revealed that BbMbp1 mediated different transcriptomes during the formation processes of conidia and blastospores. Yeast one-hybrid assays demonstrated that BbMbp1 was required for transcriptional control of a cell wall protein gene, BbCwp, and an integral membrane protein gene, BbImp that played significant roles in conidiation and blastospore formation respectively. Our results demonstrate that BbMbp1 contributes to the morphological transitions in the pathogenic and saprophytic growth of B. bassiana via different genetic pathways.

The chemical performance of 5-bromo-2-(bromoacetyl)-thiophene (1) was tested toward the reaction with numerous bi-nucleophilic reagents (namely; 2-aminobenzothiazoles, 2-aminothiazole, 2-aminotetrazole, 2-aminotriazole, 2-aminopyridines, 2-aminobenzimidazole ando-phenylenediamine). Herein, perylene diimide (PDI)/oxygen-doped g-C3N4 nanosheets (O-CN) n-n heterojunction photocatalyst was prepared by an in-situ electrostatic assembling. The pi-pi interaction between self-assembled PDI and O-CN could lead to the electron delocalization effect to facilitate the electron migration. Self-assembled PDI could broaden the visible response range of O-CN to produce more photogenerated carries. Moreover, the interlaced band structures of O-CN and self-assembled PDI could result in a built-in electric field to promote the interfacial charge separation. Besides, many active species (h(+), O-center dot(2-) and O-1(2) ) could be accumulated to improve the oxidation ability of PDI/O-CN. Therefore, PDI/O-CN exhibited enhanced visible photocatalytic oxidation activities towards disinfection, degradation and oxygen evolution. The optimum PDI/O-CN inactivated 96.6% of Staphylococcus aureus cells within 3 h, while O-CN only killed 62.2%. Additionally, the phenol degradation rate and evolved oxygen amount in 2 h of PDI/O-CN-40% were 3.6 and 1.8 times as high as those of O-CN, respectively.

Metal-organic framework compounds with abundant network structures are widely used in gas separation and storage, non-linear optics, catalysis, functional materials, and other applications. Here, self-doped nitrogen porous carbon was prepared by a one-step carbonization of Zn-2(ATRZ)(2)SO4 center dot 3H(2)O synthesized using a hydrothermal method. The self-doped nitrogen porous carbon material (ZYC-800) features a three-dimensional network structure, high specific surface area of 725 m(2) g(-1), and high nitrogen (7.26%) and oxygen (5.46%) contents. Consequently, the ZYC-800 electrode shows a high specific capacitance of 205.5 F g(-1)at a current density of 1.0 A g(-1)and high capacitance retention rate of 71.1% after 5000 cycles. Moreover, a composite electrode material (ZYC-800-5.3) with small redox additives was prepared and showed a high specific capacitance of 274.1 F g(-1)at a current density of 1.0 A g(-1). After 5000 cycles, the capacitance retention rate is 72.8%. The results show that the addition of redox additives in self-doped nitrogen porous carbon is an effective method in the development of high performance supercapacitors.

Impaired synthesis or action of thyroid hormones (THs) during critically sensitive periods of development can have long term adverse effects on health. Development of rapid assays to identify chemicals that impair THs physiology is an important goal for reducing risks from chemical use. Thyroid peroxidase (TPO) is a key enzyme regulating THs synthesis in thyroid gland and a vulnerable target for chemicals that disrupt THs synthesis. To develop a human-relevant, rapid assay for TPO inhibition, we have engineered two cell lines (CHO and LentiX-293) to express active human TPO (hTPO) enzyme and applied them in a recently-described assay using a stable fluorescent product (Amplex UltraRed). Assay performance was assessed by comparing activity of 19 reference chemicals with known strong, weak or no TPO inhibitory activity. The assay using hTPO from either cell line consistently identified the relative potency of strong to moderate inhibitors and chemicals known to be inactive. Results were less consistent for chemicals reported to be weak inhibitors of rodent TPO, possibly suggesting some species specificity. Our studies support the use of hTPO from stably transfected cell lines to substitute for animal-derived thyroid microsomes for rapid high throughput screening assays to identify and characterize TPO inhibitors.

Cationic polymers are promising antibacterial agents because bacteria have a low propensity to develop resistance against them, but they usually have low biocompatibility because of their hydrophobic moieties. Herein, we report a new biodegradable and biocompatible chitosan-derived cationic antibacterial polymer, 2,6-diamino chitosan (2,6-DAC). 2,6-DAC shows excellent broad-spectrum antimicrobial activity with minimum inhibitory concentrations (MICs) of 8-32 mu g/mL against clinically relevant and multidrug-resistant (MDR) bacteria including Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, and Acinetobacter baumannii. Furthermore, 2,6-DAC shows an excellent synergistic effect with various clinically relevant antibiotics proved by decreasing the MICs of the ntibiotics against MDR A. baumannii and methicillin-resistant Staphylococcus aureus to <1 mu g/mL. In vivo biocompatibility of 2,6-DAC is proved by a dosage of 100 mg/kg compound via oral administration and 25 mg/kg compound via intraperitoneal injection to mice; 2,6-DAC does not cause any weight loss and any significant change in liver and kidney biomarkers or the important blood electrolytes. The combinations of 2,6-DAC together with novobiocin and rifampicin show >2.4 log(10) reduction of A. baumannii in murine intraperitoneal and lung infection models. The novel chitosan derivative, 2,6-DAC, can be utilized as a biocompatible broad-spectrum cationic antimicrobial agent alone or in synergistic combination with various antibiotics.

Its efficacy affirmed in the preparation of bis (2,3-dihydroquinazolin-4(1H)-one) derivatives via one-pot pseudo five-component reaction of aldehydes, amines, and isatoic anhydride in 70 degrees C aqueous media. The protocol contains several advantages such as relatively short reaction times, simple work-up procedure, economic and environmentally-friend because of performing the two class of MCRs under green media, utilizing wide-range of substrates, and reusability and recoverability of the core-shell nano-promoter for 3 runs without significant activity loss.
