After cooling, MeOH was added, the insoluble material (copper powder) was filtered through a Celite pad, and rinsed with methanol. The filtrate was concentrated in vacuo. The residue was purified by silica gel column chromatography (50% EtOAc/CH2Cl2) to obtain 450 mg of the products as an about 4:1 mixture of two regio-isomers. This was further separated by C-18 reverse phase silica gel (YMC, ODS-A 75 mum) eluted with 15% CH3CN/H2O containing 0.1% TFA. The fractions containing the major isomer were concentrated in vacuo to remove acetonitrile and the aqueous solution was extracted with CH2C12 after neutralizing with aqueous sodium bicarbonate to obtain the title compound 3-82 (305 mg, 1.25 mmol; Y. 25%): HPLC>97% (AP at 254 nm); MS (LC/MS) m/z 244 (M+H); 1H NMR (CDCl3) delta ppm 1.43 (3H, t, J=7.5 Hz; CH3), 2.91 (2H, q, J=7.5 Hz; CH2), 4.05 (3H, s, OCH3), 6.71 (1H, dd, J=6, 2.4 Hz, H-3), 7.57 (1H, t, J=3 Hz, H-2), 7.57 (1H, s, H-5), 9.16 (1H, s, triazole-H-5), 10.3 (1H, br, NH).

To a solution of N-(3-bromo-4-fluorophenyl)-N’-hydroxy-4-nitro-l,2,5- oxadiazole-3-carboximidamide (100 mg, 0.289 mmol) in DMF (2.0 mL) at 25 C was added NaSH (25 mg, 0.446 mmol). The mixture was stirred at 25 C for 30 min. Then K2C03 (40 mg, 0.289 mmol) and 3-(chloromethyl)-lH-l,2,4-triazol-5(4H)-one (58 mg, 0.434 mmol) were added. The mixture was stirred at 25 C for 30 min, diluted with sat. NaHCC (10.0 mL, aq), and extracted with ethyl acetate (10.0 mL). The organic layer was washed with brine (5.0 mL), dried over MgSC>4, and concentrated. The residue was purified by reverse phase HPLC on a GILSON 281 instrument fitted with a Phenomenex Synergi C18 (250*21.2 mm*4 muiotaeta) using water (0.2% formic acid) and acetonitrile as eluents (Mobile phase A water(0.2% Formic acid), Mobile phase B acetonitrile, Detective wavelength: 220 nm) followed by freeze-drying to give the title compound (40 mg) as a solid. lH NMR (400 MHz, CD30D) delta 7.09 (dd, J=5.8, 2.5 Hz, 1 H), 7.04 (t, J=8.7 Hz, 1 H), 6.73 – 6.81 (m, 1 H), 4.27 (s, 2 H). ESI MS m/z 430 [M+H]+

To a stirred solution of Compound 1g in ACN (100 ml) were added Cs2CO3 (125.6 g, 386.15 mmol) and ethyl 2-bromopropanoate (39.43 ml, 283.15 mmol) at 25° C. The organic layer was dried over anhydrous sodium sulfate and concentrated to obtain 35 g crude product. The crude product was purified by normal phase column chromatography using silica (100-200 mesh), eluting with 2percent MeOH in DCM. The collected fractions were evaporated to obtain Compound 2g (30 g, 59.17percent) as a pale yellow liquid. The desired isomer was confirmed by NOE analysis. 1H NMR (400 MHz, DMSO-d6) delta ppm: 5.22-5.34 (m, 1H), 4.03-4.18 (m, 2H), 2.32 (s, 3H), 2.16 (s, 3H), 1.60 (d, J=7.23 Hz, 3H), 1.06-1.19 (m, 3H); LCMS: 75.07percent (198.17, M+H), RT=1.18 min.

5-(3-oxa-9-azabicyclo[3.3.1]nonan-7-yl)-3-(6-phenylpyridin-3-yl)-8,9- dihydropyrazolotl,5-a]pyrido[3?2-e]pyrimidin-6(7H)-one (49.78 mg, 0.44 mmol) in DMF (2 mL) was treated with EDCI (129.47 mg, 0.678 mmol) and HOBt (45.6 mg, 0.339 mmol). Then substrate 5-(3-oxa-9-azabicyclo[3.3.1]nonan-7-yl)-3-(6-phenylpyridin-3-yl)-8,9~dihydropyrazolo[l,5-a]pyrido[3,2-e]pyrimidin-6(7H)-one (122 mg, 0.21 rnmol) followed by DIEA (0.29 mL, 1.69 mmol) was added. After 30 min, the reaction mixture was treated with water (0.4 mL) and DMSO-MeCN (3:1, 3 mL). Pure compound 5-((lR,5S)-9-(lH- 1,2,4- triazole-3-carbonyl)-3-oxa-9-azabicyclo[3.3.1]nonan-7-yl)-3-(6-phenylpyridin-3-yl)-8,9- dihydropyrazolo[l,5-a]pyrido[352-e]pyrimidin-6(7H)-one was isolated by preparative HPLC. HPLC-MS 4.35 min (UV 254 r.m). Mass calculated for formula C30H27N9O3 561.2; observed MH+ (LCMS) 561.98 (m/z).

To a flask containing 1-methyl-1H-1,2,3-triazole (200 mg, 2.41 mmol) was added THF (15 mL) and the solution was cooled to -45 C. using a CH3CN-CO2 bath. n-BuLi (2.5 M in hexanes, 1.0 mL, 2.5 mmol) was added dropwise which afforded an opaque white suspension. The mixture was stirred at -45 C. for 25 minutes, then a solution of (4-chloro-3-cyclopentyl-2-methoxyquinolin-6-yl)(1,2-dimethyl-1H-imidazol-5-yl)methanone (475 mg, 1.24 mmol, Intermediate 68: step c) in THF (3 mL) was introduced. After 10 minutes, the -45 C. bath was replaced with an ice-water bath. After 30 minutes, the reaction mixture was quenched with aqueous NH4Cl solution and extracted with EtOAc:THF (2:1, 3*40 mL). The combined organics were washed with brine, dried over MgSO4, filtered and concentrated. Chromatography on silica gel (3% MeOH-DCM increasing to 10% MeOH) provided the title compound as a light tan solid. 1H NMR (500 MHz, CDCl3) delta ppm 8.24 (d, J=1.9 Hz, 1H), 7.69 (d, J=8.7 Hz, 1H), 7.35 (dd, J=8.7, 2.0 Hz, 2H), 7.07 (s, 1H), 6.01 (s, 1H), 4.07 (s, 3H), 3.90-3.80 (m, 4H containing a 3H singlet at 3.86), 3.32 (s, 3H), 2.06-1.94 (m, 3H), 1.94-1.84 (m, 6H), 1.75-1.69 (m, 2H). MS (ESI): mass calcd. for C24H27ClN6O2, 466.2. m/z found 467.1 [M+H]+. The racemate was separated by chiral SFC (Stationary phase: Chiralpak AD-H, 5 mum 250×20 mm, Mobile phase: 70% CO2: 30% EtOH (0.3% iPrNH2)) to give Example 135b as the first compound that eluted from the chiral column and Example 135c as the second compound that eluted from the chiral column.

A mixture of 3-methyl-lH-l,2,4-triazole (5.0 g, 60.2 mmol), 1- chloro-2-methoxy-4-nitrobenzene (11.3 g, 60.2 mmol), and KOH flakes (3.4 g, 48.1 mmol) in anhydrous DMSO (50 mL) was heated at 80 0C for 6 h. The reaction mixture was allowed to cool to rt and was poured into 800 mL of water with vigorous stirring. The aqueous mixture was extracted with EtOAc (3 x 200 mL). The combined organics were washed with brine, dried over Na2SO4, filtered and concentrated in vacuo. The crude residue was purified using silica gel chromatography (0-2% MeOH/chloroform, linear gradient) to afford l-(2-methoxy-4- nitrophenyl)-3-methyl-lH-l,2,4-triazole (3.7 g, 26 % yield). LC-MS (M+H)+ = 235.2.

The resulting suspension was stirred at room temp during 30 min and trideuterio(iodo)methane (319 mg, 2.2 mmol) was added and the reaction was stirred at 40C over night.The reaction mixture was poured into 50 mL H20 and extracted with EtOAc (3 x 50 mL). The organic layers were washed with sat NaCl (3 x 50 mL), dried over MgSO4 and concentrated in vacuo. The title compound was isolated as an off-white solid (456 mg, 84.8 % yield). MS (ES+) m/z: 244.9 [(M+H)+].
