Design, synthesis, DNA binding, modeling, anticancer studies and DFT calculations of Schiff bases tethering benzothiazole-1,2,3-triazole conjugates

In an attempt to design and prepare a new library of anticancer candidates, focused thiopropargylated benzothiazole was reacted with ethyl azidoacetate and/or ethyl azidobenzoate to yield newer 1,2,3-triazole-benzothiazole conjugates bearing ester functionality through click chemistry approach. The hydrazinolysis of the obtained ester-based triazoles was also carried out to give the corresponding 1,2,3-triazole acid hydrazide derivatives as precursors for the synthesis of the focused Schiff bases by their condensation with various benzaldehyde derivatives. Spectroscopic study was investigated on the establishment of the structures of all newly synthesized Schiff bases bearing benzothiazole-1,2,3-triazole molecular conjugate. The newly designed hydrazones showed two isomers ( cis-E and trans-E ) with different isomeric distribution as confirmed by NMR spectral data and supported by DFT carried out in gas phase at B3LYP 6-311G (d,p) basis set. The DFT results showed that the cis-E isomer is the lower energy structure and this finding was illustrated in terms of the intermolecular H-bonding. These molecules were screened for anticancer activities with A549 and H1299 lung cancer cell lines. The anticancer activities ranged from 55 to 90%. DNA binding study was also carried out to see the mechanism of action and the DNA binding constants were of good value ranging from of 2.0 x 10(5) and 14.7 x 10(5) M-1; indicating good interactions of the reported molecules with DNA. Finally, the modeling was confirmed and it was found that the results of modeling were in good agreement with the results of anticancer and DNA binding studies. Tripodal structured blue-green emissive fluorescent sensors for highly selective bifunctional detection: Their logic gate operations and real sample applications

The three novel tripodal-structured polyaromatic hydrocarbon-based fluorescent sensors which contain triazole as binding sites and naphthalene, anthracene, pyrene moieties as fluorophores (TN, TA, and TP) were synthesized via click chemistry and fully characterized by standard spectroscopic techniques. The photophysical and fluorescent sensor properties of TN, TA, and TP were investigated by UV-Vis electronic absorption, steady-state/time resolved/3-D fluorescence, and EEM-analysis spectroscopies. It was found that fluorescent sensors demonstrated excellent sensitivity and selectivity for Fe3+ ions over-tested other metal ions with significant fluorescence turn-off’ responses which led to instant naked-eye and fluorescence color change. The binding mechanisms of TN, TA, and TP for F were evaluated with Job’s plot and mass analysis. Moreover, treatment with H led to a sharp be increased in fluorescence intensities of TN, TA, and TP with turn-on fluorescence responses of Fe complexes ensembles. The obtained LODs (0.25-0.74 mu M) for iron ions were significantly lower than the tolerated limit of iron for drinking water by WHO/US EPA. The highly selective, time-saving, and sensitive fluorescence turn-off’ responses of fluorescent sensors for Fe3+ ions were used for iron determination in environmental water samples. In addition, reversible fluorescence quenching responses of presented fluorescent sensors after addition of H were utilized for improvement of IMPLICATION logic gates.

Cross-resistance to the new fungicide mefentrifluconazole in DMI-resistant fungal pathogens

In the European Union (EU), regulation of sterol demethylation inhibiting (DMI) fungicides is tightened due to their suspected endocrine disrupting properties. However, the new DMI fungicide mefentrifluconazole was reported to have high fungicidal activity with minimal adverse side effects. In addition, some evidence suggests inconsistent cross resistance between mefentrifluconazole and other azoles. In this study, mefentrifluconazole and other triazoles were examined for activity to select pathogens sensitive or resistant to DMIs using mycelial growth tests on fungicide-treated culture medium or spray trials using cucumber plants. Cross-resistance was confirmed for all of the fungal species tested but activity levels varied. The sensitivity of Monilinia fructicola from peach to mefentrifluconazole was higher compared to other DMIs. In contrast, the inhibitory activity of mefentrifluconazole was equal or slightly inferior compared to difenoconazole, tebuconazole, propiconazole in Colletotrichum spp., Alternaria alternaria sp. complex and Cercospora beticola isolated from peach and sugar beet, respectively. Similar tendencies (i.e. equal or slightly inferior activity and cross-resistance) were observed for cucumber powdery mildew (Podosphaera xanthii) resistant to triflumizole, myclobutanil, and difenoconazole. Despite cross-resistance to other DMIs, mefentrifluconazole is a promising fungicide for fungal disease control on peach and other crops, with a reportedly more favorable toxicity profile.

Pesticide multi-residues in Dendrobium officinale Kimura et Migo: Method validation, residue levels and dietary exposure risk assessment

Dendrobium officinale Kimura et Migo (D. officinale) is a dual-use plant with both botanical medicine and food applications, drawing increasing attentions. Pesticides are inevitably applied on D. officinale in commercial artificial-sheltered cultivation, yet little is known about pesticide residue levels in D. officinale. A modified high through-put QuEChERS method coupled with HPLC-MS/MS was developed and validated to detect 76 pesticides in D. officinale. Graphitized multi-wall carbon nanotubes (g-MWCNTs) was selected as the clean-up sorbent, showing relative weak affinity to triazole fungicide having planar structure in their molecular and low matrix effects of pesticides in D. officinale samples compared to MWCNTs and pesticarb. The validated method was applied to analyze pesticide residues in 86 real D. officinale samples collected from three main producing provinces. 43 different pesticides were detected with highest residue of 6.11 mg/kg for dimethomorph. Given possible health risks related to pesticide residues, accordingly, risk assessment of human exposure to pesticides via intake of D. officinale was thus performed, indicating that the pesticide residue in fresh or dry D. officinale would not cause potential risk to human health either in the long-term or short-term. This work improved our understanding of potential exposure risk of pesticide multi-residues in D. officinale.

A novel catalyst is designed and synthesized based on immobilization of copper onto modified magnetic nanoparticles. The catalyst was characterized by several characterization techniques. The catalyst was applied for the synthesis of a novel series of heterocyclic scaffold containing a 1,4-benzodiazepine fused with a 1,2,3-triazole ring and a quinazolin-11(9H)-one skeleton. The method is based on the direct synthesis of quinazolin-11(9H)-one ring by the reaction of isatoic anhydride (or anthranilic acid) and propargylamine, and an aldehyde, containing a leaving group on its aromatic ring. The reaction contains an in situ sequential click reaction of azide group with terminal alkyne and subsequent N-arylation reaction to afford the final products. The method benefits various advantages, such as regioselectivity and high yield of the products. Developmental reproductive toxicity and endocrine activity of propiconazole in the Xenopus tropicalis model

Environmental pollutants and especially endocrine disrupting chemicals (EDCs) are implicated as one of the drivers of the amphibian declines. To advance the understanding of the risks of EDCs to amphibians, methods to determine endocrine-linked adverse effects are needed. The aims were to 1) develop a partial life-cycle assaywith themodel frog Xenopus tropicalis to determine endocrine perturbation and adverse developmental effects, and 2) determine effects of propiconazole in this assay. Propiconazole is a pesticidewithmultiple endocrine modes of action in vitro. Its potential endocrine activity and adverse effects in amphibians remain to be elucidated. Tadpoles were exposed to 0, 33 and 384 mu g propiconazole/L during critical developmental windows until completed metamorphosis. At metamorphosis, a sub-sample of animals was analysed for endpoints for disruption of estrogen/androgen (sex ratio, brain aromatase activity) and thyroid pathways (time to metamorphosis). The remaining individuals were kept unexposed for 2 months post-metamorphosis to analyze effects on sexual development including gonadal and Mullerian duct maturity and gametogenesis. At metamorphosis, brain aromatase activity was significantly increased in the high-dose group compared to control. In both propiconazole groups, an increased proportion of individuals reached metamorphosis faster than the mean time for controls, suggesting a stimulatory effect on the thyroid system. At 2 months post-metamorphosis, testis size, sperm and Mullerian duct maturity were reduced in the low-dose males, and the liver somatic index in males was increased in both propiconazole groups, compared with controls. In conclusion, our results show that propiconazole exposure caused endocrine perturbations and subsequent hepatic and reproductive effects evident at puberty, indicating persistent disruption of metabolism and male reproductive function. Our findings advance the development of methodology to determine endocrine and adverse effects of EDCs. Moreover, they increase the understanding of endocrine perturbations and consequent risk of adverse effects of azoles in amphibians. (C) 2020 The Authors. Published by Elsevier B.V.

It was found that 1 can be as a heterogeneous catalyst for multiple organic reactions, such as azide-alkyne cycloaddition and Friedel-Crafts reactions with good isolated yields and good recycle runs (at least five times without substantial degradation).
